| 摘 要： | 目的观察存活蛋白反义寡核苷酸转染对食管癌细胞凋亡、增殖、对化疗药物敏感性的影响。方法设计合成特异性存活蛋白反义寡核苷酸（ASODN）。食管癌EC109细胞系分为6组：空白对照组、空脂质体转染对照组、正义链转染对照组、160、200、240nmol／L ASODN转染组。作用20h后收获各组细胞，Western blot法检测各组细胞生存素表达情况。TUNEL法检测各组细胞的凋亡情况。结果各ASODN转染组细胞存活蛋白表达有不同程度减弱，细胞变圆、折光增强、细胞碎片形成等，而各对照组细胞生长良好。各ASDON转染组细胞凋亡指数明显高于各对照组（P〈0．05），以240nmol／L转染组最明显（P〈0．05）。结论封闭食管癌细胞存活蛋白基因，能下调存活蛋白的表达，从而诱导食管癌细胞凋亡，抑制食管癌的发展。

| Abstract: | Objective To investigate the effects of antisense oligonucleotide(ASODN) targeting survivin on esophageal carcinoma cell apoptosis and sensitivity to chemotherapeutic drugs.Methods ASODN targeting survivin was designed and constructed,and it was transfected by Oligofectamine.Survivin protein expression was detected by Western Blot;apoptotic index(AI) was examined by TUNEL.Results Expression of Survivin in ASODN groups were significantly decreased compared with that in control groups;the AI of ASODN groups was significantly higher than that of other groups.Conclusion Expression of survivin may decrease in EC109 cells after ASODN transfection.ASODN targeting Survivin can induce apoptosis in esophageal carcinoma. |
