结直肠癌中肿瘤相关巨噬细胞及基因表型的分析研究

目的结直肠癌(Colorectal cancer,CRC)中肿瘤相关巨噬细胞(TAMs)CD68~+及CD163~+的检测及与基因表型的关系。方法免疫组化方法检测41例CRC肿物,癌旁5 cm(M5)、10 cm(M10)。不典型增生(Atypical hyperplasia,AH)组织CD68~+及CD163~+的表达,采用病理切片扫描仪细胞计数。对41例CRC肿物进行基因检测。结果CRC患者CD68~+及CD163~+检测肿物组织高于癌旁5 cm、10 cm(P0.01);肿物与不典型组织比较差异无统计学意义(P0.05);癌旁组织M5与M10比较差异无统计学意义(P0.05);回归分析提示CD163~+表达与K-ras基因突变有显著性关系(P0.05)。CD68~+及CD163~+的表达与B-raf基因突变、MSS(微卫星稳定型)、淋巴结转移、脉管癌栓、神经侵犯无相关性(P0.05)。结论 CD68~+及CD163~+在结直肠癌肿物间质中高表达,并高于癌旁组织,与AH表达无相关性。CD163~+在肿物的表达与K-ras基因突变有相关性。

Analysis of tumor-associated macrophages and gene phenotypes in colorectal cancer

Objective To detect the expression of CD68+and CD163+in tumor-associated macrophages(TAMs)in colorectal cancer(CRC)and its relationship with gene phenotype.Methods Immunohistochemical method was used to detect the expression of CD68+and CD163+in 41 cases of CRC,paracancer 5 cm(M5),10 cm(M10),and atypical hyperplasia(AH)tissues.Cell counts were performed using a pathological slice scanner.Gene detection was performed on 41 CRC tumors.Results The expression of CD68+and CD163+in CRC patients was higher than that of paracancer 5 cm and 10 cm tissues(P<0.01).There was no significant difference between tumor and atypical tissue(P>0.05).The difference was not statistically significant between paracancer M5 and M10 tissues(P>0.05).Regression analysis indicated that CD163+expression was significantly associated with K-ras gene mutation(P<0.05).The expression of CD68+and CD163+was not associated with B-raf gene mutation,MSS(microsatellite stable),lymph node metastasis,vascular tumor thrombus,and neurological invasion(P>0.05).Conclusion CD68+and CD163+are highly expressed in the interstitial of colorectal cancer,and higher than the adjacent tissues,and have no correlation with AH expression.The expression of CD163+in the tumor is correlated with the K-ras gene mutation.
