摘    要:目的比较不同产地穿山龙Dioscorea nipponica Makino中薯蓣皂苷元的含有量。方法采用加压液体萃取法获得薯蓣总皂苷,并利用加压两相酸水解法制备供试品溶液。穿山龙50%乙醇提取物的分析采用Waters SunFire RP18柱(150 mm×4.6 mm,5μm);流动相乙腈-水;体积流量1.0 mL/min;检测波长203 nm;柱温30℃。结果通过新方法制备所得的供试品溶液中薯蓣皂苷元的平均含有量比传统方法高出18%。薯蓣皂苷元在10.94~1 400μg/mL范围内线性关系良好(r=0.999 5),平均加样回收率96.88%~103.0%,RSD<3%,平均含有量0.695%~2.263%。结论不同产地穿山龙中薯蓣皂苷元的含有量差异很大,其中产于黑龙江伊春的最高,且东北三省产的远高于其他产地。

Abstract:AIM To compare the content of diosgenin in Dioscorea nipponica Makino from different growing areas.METHODS Pressurized liquid extraction was performed to obtain total dioscin, followed by biphasic acidic hydrolysis to prepare test solution. The analysis of 50% ethanol extract of D. nipponica was performed on a 30 ℃ thermostatic Waters SunFire RP18 column(150 mm×4.6 mm, 5 μm), with the mobile phase comprising of acetonitrile-water flowing at 1.0 mL/min,and the detection wavelength was set at 203 nm.RESULTS Compared to conventional method, the content of diosgenin in test solution was 18% higher by the novel method. The diosgenin constituents showed good linear relationships within 10.94-1 400 μg/mL(r=0.999 5), whose average recoveries were 96.88%-103.0% with the RSD<3%.The content of diosgenin in D. nipponica was 0.695%-2.263%. CONCLUSION The content of diosgenin is quite different in D. nipponica tuber from different growing areas. The samples collected from Yichun city Heilongjiang province have the highest content of diosgenin, and the samples from three Northeast provinces of China have higher content of diosgenin than others.
