黄芪总黄酮对脑缺血再灌注损伤大鼠氧化应激、炎症、凋亡的影响

摘    要:目的探讨黄芪总黄酮对脑缺血再灌注损伤大鼠氧化应激、炎症、凋亡的影响。方法 SD大鼠随机分为假手术组、模型组、尼莫地平(15 mg/kg)组及黄芪总黄酮低、高剂量组(30、60 mg/kg),灌胃给药15 d后线栓法制备脑缺血模型(缺血2 h后再灌注24 h)。然后,计算神经功能学评分、脑组织含水量,ELISA法检测MDA水平和GSH-Px、SOD活性,放射免疫法检测IL-1β、TNF-α水平,实时荧光定量PCR法检测caspase-3、Bcl-2、Bax mRNA表达,Western blot法检测p-PI3K、p-AKT蛋白表达。结果与模型组比较,黄芪总黄酮组神经行为学评分,脑组织含水量,MDA、IL-1β、TNF-α水平,caspase-3、Bax mRNA表达显著下降(P<0.05),GSH-Px、SOD活性,Bcl-2 mRNA表达,p-PI3K、p-AKT蛋白表达显著升高(P<0.05)。结论黄芪总黄酮对脑缺血再灌注损伤大鼠具有明显的保护作用,其机制可能与抑制氧化应激水平、减少炎症反应、抗凋亡作用有关。

Abstract:AIM To study the effects of Astragali Radix total flavonoids on oxidative stress, inflammation and apoptosis of rats with cerebral ischemia-reperfusion injury.METHODS SD rats were randomly divided into sham-operation group, model group, nimodipine(15 mg/kg) group and Astragali Radix total flavonoids high-dose(60 mg/kg), low-dose(30 mg/kg) groups, thread thrombus method was applied to preparing cerebral ischemia model(24 h of reperfusion after 2 h of ischemia) after 15-day intragastric administration. Subsequently, neurological score and water content in brain tissue were calculated, MDA level and GSH-Px, SOD activities were detected by ELISA, IL-1β and TNF-α levels were detected by radioimmunoassay, caspase-3, Bcl-2 and Bax mRNA expressions were detected by RT-PCR, p-PI3 K and p-AKT protein expressions were detected by Western blot.RESULTS Compared with the model group, the Astragali Radix total flavonoids groups demonstrated significantly decreased neurological score, water content in brain tissue, MDA, IL-1β and TNF-α levels, caspase-3 and Bax mRNA expressions(P<0.05), and markedly increased GSH-Px and SOD activities, Bcl-2 mRNA expression, p-PI3 K and p-AKT protein expressions(P<0.05).CONCLUSION Astragali Radix total flavonoids exhibit obvious protective effects on rats with cerebral ischemia-reperfusion injury, whose mechanisms may contribute to the inhibition of oxidative stress levels, reduction of inflammatory reactions and anti-apoptosis effect.
