淫羊藿苷脂质体经鼻给药对帕金森病大鼠行为学及神经递质的影响

摘    要:目的观察经鼻给予淫羊藿苷脂质体对帕金森病模型大鼠的行为学症状及纹状体相关神经递质含量的影响。方法按随机原则选16只大鼠为假手术组(假手术/in组),另将64只造模成功的PD大鼠分为PD模型组(PD/in组)、淫羊藿苷脂质体经鼻给药组(ICA-PGL/in组)、淫羊藿苷经鼻给药组(ICA/in组)、淫羊藿苷脂质体静脉注射给药组(ICA-PGL/iv组),每组16只。药物持续干预2周后,通过检测大鼠阿扑吗啡旋转次数、圆筒实验、纹状体区TH表达以及相关神经递质含量的改变来观察淫羊藿苷脂质体结合经鼻给药方式对帕金森病模型大鼠的疗效。结果淫羊藿苷各治疗组运动功能较PD/in组有不同程度的改善,纹状体区双侧TH阳性纤维表达及纹状体区右侧单胺类神经递质DA、DOPAC和HVA含量较PD/in组均有不同程度的提高,其中以ICA-PGL/in组疗效最为突出(P0.01)。结论经鼻给药淫羊藿苷脂质体对帕金森病模型大鼠运动功能及神经递质的修复有不错的疗效。

Effect of intranasal administration of icariin liposome on behavior and neurotransmitter in rats with Parkinson's disease
Abstract:Objective To observe the effects of nasal administration of icariin liposome on behavioral symptoms and striatum-associated neurotransmitter levels in Parkinson''s disease model rats.Methods Sixteen rats were randomly selected as sham operation group(sham operation/in group),and 64 successful modeling PD rats were divided into PD model group(PD/in group)and icariin liposome intranasal administration group(ICA-PGL/in group),icariin intranasal administration group(ICA/in group),and icariin liposome intravenous administration group(ICA-PGL/iv group),16 rats in each group.After two weeks of continuous drug intervention,the efficacy of icariin liposome combined with nasal administration on Parkinson''s disease model rats was observed by numbers of apomorphine rotation,cylinder test,TH expression in striatum and changes of related neurotransmitters.Results The results showed that the motor function of each icariin treatment group was improved to some extent when compared with the PD/in group.The expression of bilateral TH-positive fibers in the striatum and the levels of monoamine neurotransmitter DA,DOPAC and HVA on the right side of the striatum were increased in different degrees when compared with PD/in group,and the effect in ICAPGL/in group was the most significant(P<0.01).Conclusion The intranasal administration of icariin liposomes has a good effect on the motor function and neurotransmitter repair in Parkinson''s disease model rats.
