4种异黄酮改构化合物对人离体子宫内膜腺上皮细胞增殖活性的比较

摘    要:目的观察4种异黄酮改构化合物(F8、F11、ZF3和ZF7)对人离体子宫内膜腺上皮细胞增殖的影响。方法采用胶原酶消化十二次筛网过滤法原代培养人子宫内膜腺上皮细胞。MTT法检测并比较4种化合物对人离体子宫内膜腺上皮细胞增殖的影响。结果成功培养原代人离体子宫内膜腺上皮细胞并稳定传代。4种异黄酮化合物作用于人子宫内膜腺上皮细胞24h后,低剂量(25μmol/L)F8显著促进细胞增殖(P〈0.05),中高剂量(50、100μmol/L)F11显著抑制细胞增殖(P〈0.05);作用48h后,中剂量(50μmol/L)F11和ZF3显著抑制细胞增殖(P〈0.05),高剂量(100μmol/L)化合物均显著抑制细胞增殖(P〈0.05);作用72h后,除低剂量(25μmol/L)F8、ZF3和ZF7未明显抑制细胞增殖外(P〉0.05),各化合物在不同剂量下均抑制细胞增殖效应(P〈0.05);该生物学效应具有时间和剂量依赖性。结论4种异黄酮化合物在一定剂量下作用一段时间后可抑制人离体子宫内膜腺上皮增殖。F11抑制细胞增殖活性最强。
