骨肉瘤中细胞增殖和细胞凋亡的关系及临床意义

目的:探讨骨肉瘤组织中细胞凋亡与细胞增殖的关系及临床意义。方法:采用原位末端转移酶法和免疫组织化学技术分别检测骨肉瘤和良性骨瘤中凋亡细胞密度和增殖细胞核抗原(PCNA)的表达。结果:4种亚型骨肉瘤组织中凋亡细胞密度均明显低于良性骨肿瘤组织,而PCNA阳性细胞密度明显高于良性骨肿瘤组织;在骨肉瘤4种亚型中凋亡细胞密度和PCNA阳性细胞密度呈明显负相关,而在良性骨肿瘤中两者呈线性正相关。结论:在骨肉瘤的发生中可能普遍存在细胞凋亡机制的紊乱可能以低凋亡高增殖表现为多,并认为细胞凋亡检测尚难以作为一种独立的预后判断指标。

RELATIONSHIP BETWEEN CELL PROLIFERATION AND APOPTOSIS AND THEIR CLINICAL SIGNIFICANCE IN OSTEOSARCOMAS

Objective: To investigate relationship between cell proliferation and apoptosis and their clinical significance in osteosarcomas. Methods: In situ terminal deoxynucleotide transferase labeling was used for detecting apoptotic cells, and immunohistochemistry for proliferating cell nuclear antigen (PCNA) in.65 osteosarcomas and 19 benign osteomas. Results: The densities of apoptotic cells in 4 subtypes of osteosarcomas was significantly lower than that in benign osteomas, and the densities of positive cells for PCNA was significantly higher. Significantly negative correlation was found between the densities of apoptotic cells and proliferation cells in osteosarcomas, and Significantly positive correlation in benign osteomas(r=0.4251). Conclusion: The data suggest that mechanism disturbance of apoptosis may be a common event in osteosarcoma genesis, and the high density of proliferative cells and low density of apoptotic cells occur more usually in osteosarcoma tissues. Predictive value of apoptosis as an independent prognostic parameters remains to be established in a larger series.
