大鼠Walker-256皮下移植瘤模型的建立及其超声评价

摘    要:目的 探讨建立大鼠Walker-256皮下移植瘤模型两种方法的有效性及三维超声在监测肿瘤生长特性中的价值。方法 24只SD大鼠随机分两组,每组12只,分别采用浓缩腹水注射法和肿瘤组织块皮下包埋法建立皮下肿瘤模型,三维容积探头观察成瘤率、肿块大小及声像图特征。三维能量多普勒显像(3D—PDI)反映两组移植瘤血管指数(VI)的变化。结果 浓缩腹水注射组接种成功率100%,高于组织块皮下包埋组(75%,P〈0.05),且肿瘤生长速度快于组织块皮下包埋组。浓缩腹水注射组和组织块皮下包埋组建立的移植瘤血管指数分别于16d、20d时达峰值。结论两种方法均可有效地建立皮下移植瘤模型,浓缩腹水注射法成瘤率高于组织块皮下包埋法。三维超声可作为监测肿瘤体积及血流的有效手段。

Abstract:Objective To investigate two methods of making rat model bearing Walker-256 subcutaneous implanted tumor and to equally the application of three-dimensional ultrasound in the characterization of subcutaneous tumor.Methods Twenty-four SD rats were randomly divided into two groups.The subcutaneous transplantation tumor model were prepared by directly injection of enriched cancerous ascites or by the implantation of tumor tissue. The positive rate of inoculation,tumor size,growth rate were observed by ultrasound volume probe.The vascularization index(VI) of two groups were subjected to three-dimensional power Doppler imaging(3DPDI) examinations.Results The positive rate of inoculation in the injection group was higher than that in the implantation group,which were 100% and 75% respectively(P<0.05),and the growth rate was faster in the injection group thanin the implantation group.The time of peak amplitude of VI of the injection group and the implantation group were 12 days and 16 days respectively.Conclusion Both the injection and the implantation technique can prepare subcutaneous transplantation tumor model in rats,but the positive rate of inoculation of the injection group was higher than that of the implantation group. Three-dimensional sonography was useful in evaluation of subcutaneous tumors.
