摘    要:目的 研究Notch1(ICN)基因对人宫颈癌细胞生长的影响,并对其作用机制进行探讨。方法 采用脂质体法转染人宫颈癌HeLa细胞,MTT比色法测定瞬时表达Notch1(ICN)对细胞生长的影响,用流式细胞仪分析细胞周期分布的变化,Western blot法检测P53蛋白的表达改变。结果 瞬时表达Notch1(ICN)基因可以抑制HeLa细胞的生长,使细胞周期停滞在G1期,并上调细胞周期调控蛋白P53的表达水平。结论 通过影响细胞周期的分布及细胞周期调控蛋白的表达,激活Notch1基因可抑制人宫颈癌细胞的生长。

Abstract:Objective To investigate the effect of Notchl signal activation on the growth of human cervical cancer cells, and to explore the related mechanisms. Methods HeLa cells with LipofectAMINE reagent were transfected. Proliferation of HeLa cells was observed by MTT assay. Cell cycle distribution was analyzed by FACS and P53 expresssion was detected by Western blot. Results After transfection, the growth of HeLa cells was inhibited significantly and FACS analysis showed that HeLa cells in G1 phase were increased. Western blot showed that Notch1 signaling up-regulated p53 expression. Conclusions Notchl signal activation could inhibit the growth of HeLa cells through cycle arrest and p53 induction.
