Reproduction of a model of homologous bladder transplantation in rats

Abstract:Objective:To reproduce a model of rat homologous bladder transplantation without blood vessel anastomosis, and to provide the experimental basis for organ transplantation. Methods.. Thirty newborn Sprague. Dawley rats were used as donors. Twenty adult 5-week-old Wistar rats underwent homologous bladder transplantation, the grafts were harvested from the newborn Sprague-Dawley rats. They were wrapped with the greater omentum of 5-week-old Wistar rats without blood vessel anastomosis. Immunosuppressive agent was used after transplantation. They were randomly divided into two groups.. 10 rats were sacrificed at the 7th day (7-day group), and 10 rats were sacrificed on the 14th day after bladder transplantation (10-day group). The survival rate of transplanted bladder grafts was observed, and the quantity of blood vessels in the bladder wall, and total area of blood vessel sections, the depth of transitional epithelium, and the thickness of muscle were determined. Results.. The rats in 7-day group all survived, and 1 rat died in 14-day group. The successful rate of the operation reached 95 %. Microscopically, it was observed that the epithelium and muscle layer and blood vessel of the grafts were in good condition. The quantity of blood vessels in the grafts were increased (P〈0.05). The total sectional area of blood vessel in the grafts was increased too (P〈0.05). There was no obvious rejection reaction in the transplanted bladder grafts. Conclusion: This model of bladder transplantation could be considered to be a kind of stable animal model. It can be reproduced in any laboratory.
