雷公藤多甙对人急性髓性白血病细胞株HL-60的生长抑制及凋亡诱导作用

摘    要:目的:探讨雷公藤多甙对人急性髓性白血病细胞株HL-60细胞的增殖及凋亡的影响。方法:应用四甲基偶氮唑蓝(MTT)法、荧光染色法、流式细胞仪(FCM)检测、DNA片段化分析等方法研究雷公藤多甙对HL-60细胞的影响。结果:雷公藤多甙能显著抑制白血病细胞的增殖,降低肿瘤细胞的存活率,其半数细胞抑制剂量( IC50 )为5.0 μg/ml,阻滞细胞周期于G2/M期(P<0.05)。雷公藤多甙实验组在形态学上可见明显凋亡细胞,且凋亡细胞百分率显著上升(P<0.05),凝胶电泳分析有DNA梯状条带出现。结论:雷公藤多甙能显著抑制人急性髓性白血病细胞株HL-60细胞的生长增殖并促进细胞凋亡。

Abstract:OBJECTIVE: To determine the effects of the glucosidorum tripterygll totorum on the proliferation inhibition and apoptosis induction of cell line HL-60 in human acute promyelocytic leukemia. METHODS: The survival rates of cells were tested by MTT assay, fluorescence stain, flow cytometry test, and DNA fragmentation. RESULTS: Glucosidorum tripterygll totorum could notably reduce the survival rate of HL-60 cells, and inhibit the proliferation of tumor cells. The 50% inhibitory concentration (IC50) was 5.0 microg/ml and it arrested the cell cycle at G2/M phase (P < 0. 05). The typical apoptosis morphologic cells were seen, the rates of apoptosis cells were increased (P < 0.05), and DNA ladder appeared in the experiment group. CONCLUSION: Glucosidorum tripterygll totorum could notably inhibit the proliferation and induce the apoptosis of HL-60 cells.
