兔腰椎间盘髓核细胞的培养及形态观察

目的：通过对兔腰椎间盘髓核细胞的培养，观察细胞内的演变，探讨髓核细胞的生物学行为及影响因素。方法：取兔腰椎间盘髓核细胞，在加10％灭活胎牛血清的F12－DMEM液中培养，建立体外髓核细胞培养模型。通过光镜、电镜观察，同时进行细胞活力测定。结果：（1）原代细胞生物学性状最接近体内细胞，传代后细胞呈现衰老现象。（2） 活力测定提示随着培养时间的延长及传代，活力逐渐降低。（3）髓核细胞内细胞器的变化与其生物学活性变化相一致。结论：兔腰椎髓核细胞的体外培养成功为人腰椎髓核细胞的培养奠定了基础。

Cells Culture of Rabbit Lumbar Nucleus Pulposus and Its Morphological Observation

Objective:To observe the continuous changes and explore the biological action and affecting factors of nucleus pulposus cells. Methods:Lumbar nucleus pulposus cells were cultured in F12 DMEM medium and 10% fetal bovine serum. We want to set up a cells culturing model,observe them through light microscope and electron microscope and assess the cells viability. Results: The biological characteristics of the primary generation of the nucleus pulposus cells were similar to those of the cells in vivo. After subculture, the cells are becoming senility. The assessment of the cell viability shows that the assessment are declined as the time of the culture passed. The structures of the organellae in nucleus pulposus cells varied with the changes of cell biological characteristics. Conclusion:This study is a basic work for culturing human degenerative nucleus pulposus cells.
