西红花酸和西红花苷对缺血缺氧损伤心肌保护作用的比较研究

摘    要:目的:比较西红花酸(Crocetin)和西红花苷(Crocin)对损伤心肌保护作用的差异。方法:用血清药理学的方法,测定细胞培养液中LDH、CK、MDA,SOD的含量及细胞活力 同时,采用大鼠冠脉左前降支结扎心肌缺血模型,观察心电图变化,检测LDH、CK,心肌梗死面积及心肌组织病理变化。结果:西红花酸含药血清能显著降低血清和缺糖缺氧心肌细胞上清中LDH、CK、MDA的含量,提高SOD的含量,增强细胞的活力,ig西红花酸能降低冠脉结扎大鼠ECG的J点位移和心肌梗死面积百分率,减轻心肌组织病理改变 而西红花苷无此作用。结论:ig西红花酸均能明显减轻心肌损伤,治疗心肌缺血,西红花酸临床给药可以口服,而西红花苷治疗心肌缺血口服是一个不合适的给药途径。

Comparison of the Effects of Crocetin and Crocin on Myocardial Injury in Rats

Abstract:AIM: To compare the effects of crocetin and crocin on myocardial injury in rats. METHODS: In vitro, the myocardial ceils injured by low glucose and hypoxia were treated with serum from rats given crocetin (50 mg.kg-1)or crocin(150 mg.kg-1). The levels of LDH ,CK ,MDA and SOD in culture medium were measured and the cell activity was detected by MTT. In vivo, a rat myocardial ischemia model was set up using the coronary ligation technique. J point shift and T wave change in the elec- trocardiograph (ECG) in 6 hours after the treatment and area of myocardial infarction were assessed. The histopathological change of myocardium was observed by HE staining. The serum samples were collected to measure the levels of LDH and CK. RESULTS: In vitro, serum containing crocetin decreased the levels of LDH ,CK and MDA, increased the level of SOD and the activity of cardiac myocytes. However, serum containing crocin showed no significant protective effect. Compared to the model group, crocetin downshified the J point and T wave in the ECG evaluation, decreased the levels of serum LDH and CK and the area of the myocardial infarction. Crocetin reduced the histopathological changes caused by myocardial injury. CONCLUSION: ig crocetin can significantly protect the injured myocardial cell both in vivo and in vitro while ig crocin can not. So, it is suggested that crocetin by oral admini- stration is suitable to cure myocardial ischemia in clinical practice, while crocin by oral administration is not.
