MMP2及MMP9在膀胱癌中的表达及在侵袭转移中的作用

目的探讨ＭＭＰ２及ＭＭＰ９在膀胱癌中的表达及在侵袭转移中的作用。方法采用原位杂交方法观察人膀胱移行细胞癌（ＴＣＣＢ）ＭＭＰ２、ＭＭＰ９、ＭＴＭＭＰ１、ＴｉＭＰ１、ＴｉＭＰ２ｍＲＮＡ表达。结果ＴＣＣＢ组织中ＭＭＰ２、ＭＭＰ９、ＭＴＭＭＰ１、ＴｉＭＰ１及ＴｉＭＰ２表达显著高于正常膀胱上皮，其中ＭＭＰ２及ＭＭＰ９表达增加较ＴｉＭＰ２及ＴｉＭＰ１更显著。阳性表达细胞在肿瘤间质界面较多。随分级升高，ＭＭＰ２、ＭＭＰ９及ＭＴＭＭＰ１表达呈增加趋势，而ＴｉＭＰ１及ＴｉＭＰ２表达呈下降趋势。死亡患者ＭＭＰ２、ＭＭＰ９、ＭＴＭＭＰ１表达有显著升高，而ＴｉＭＰ１及ＴｉＭＰ２有显著下降。结论ＭＭＰ２及ＭＭＰ９参与ＴＣＣＢ侵袭和转移，具有较高的预后价值。

The expression of MMP 2 and MMP 9 in transitional cell carcinoma of bladder and its correlation to the tumor invasion and metastasis

Objective To elucidate the effect of mRNA expression of MMP 2,MMP 9,MT MMP 1,TiMP 1 and TiMP 2 on transitional cell carcinoma of bladder (TCCB). Methods 25 human TCCB and 5 normal bladder paraffin embedded tissue were studied by in situ hybridization mRNA. Results (1) In TCCB, the mRNA expression of MMP 2, MMP 9, MT MMP 1, TiMP 1 and TiMP 2 were increased significantly as comparing to normal bladder epithelial and the enhancement of MMP 2 and MMP 9 was greater than that of TiMP 1 and TiMP 2. The positive expression tumor cells tended to gather in the tumor stroma interface. These suggested that the MMP 2 and MMP 9 were important to Tccb invasion and metastasis. (2) The expression of MMP 2, MMP 9 and MT MMP 1 tended to increase, With the tumor grade while the expression of TiMP 1 and TiMP 2 tended to decrease. In deadly cases, the expression of MMP 2, MMP 9 and MT MMP 1 was increased significantly, while the expression of TiMP 1 and TiMP 2 was decreased significantly. In recurrent patients the expression of above mentioned mRNA was not significantly different to that of patients who were free of tumor for more than 5 years.Conclasions These suggested that the expression of MMP 2 and MMP 9 was useful in TCCB prognosis.
