白癜风患者血清抗黑素细胞抗体与恶性黑素瘤的相关性研究

目的分析白癜风患者血清抗黑素细胞抗体与恶性黑素瘤的相关性。方法利用活黑素细胞ELISA法筛选白癜风患者血清;制备黑素瘤细胞的全息膜蛋白,通过双向电泳技术分离其蛋白;转膜后分别用白癜风患者抗黑素细胞抗体阳性血清和正常血清作为一抗行W estern B lotting检测。结果获得抗黑素细胞抗体的阳性血清;优化膜蛋白提纯的条件,获得了黑素瘤细胞全息膜蛋白分辨率高、重复性好的双向电泳图;利用W estern B lotting方法比较正常人血清和患者血清与黑素瘤细胞膜蛋白的免疫印迹点,发现1个阳性差异蛋白点,表观分子量为70-75KD,表观等电点约为6.5。结论证明了在黑素瘤细胞表面存在与白癜风患者血清抗黑素细胞抗体结合的膜抗原。

Study of the Relevance of Anti-melanocyte Antibody in Vitiligo and Melanoma

Objective To study the relevance of anti - melanocyte antibody in vitiligo and melanoma. Methods Sera from patients with vitiligo were screened by using live melanocyte cell enzymelinked immunoabsorbent assay;Prepare membrane proteins of melanoma cells, separate proteins by 2-DE ;Then,the proteins were transferred from two dimensional gel onto nitrocellulose membrane,followed by incubation with sera from people with and without vitiligo,to confirm whether there were the anti-melanocyte antibody in vitiligo binding to the melanoma cell membrane proteins. Results We acquired the sera of anti - melanocyte antibody and the satisfied two dimensional gel images of proteins extracted from melanoma membranes;Through Western Blotting technique, we found that there was the one differently dot between the two cell membranes,that apparent molecular weight about 70 -75KD, the isoelectric point about 6.5. Conclusion To prove there are the anti-melanocyte antibody in vitiligo binding to the melanoma cell membrane proteins.
