| 摘 要： | 尿微量白蛋白的检测对肾损伤的早期诊断及治疗具有重要意义，表面等离子体谐振生物传感器具有无需标记、灵敏度高的特点，因此有望在本领域发挥重要作用。本研究利用自制的SPR生物传感器，对检测尿微量白蛋白的方法进行了研究。同时利用直接法和间接法对人血清白蛋白进行检测，并对两种方法进行了比较。直接法可以检测到0．1ug／mL的人血清白蛋白，问接法的检测范围为1—50ug／mL，两种方法分别可通过双抗夹心法或二抗对传感器响应值进行放大，以进一步提高灵敏度。

| Abstract: | The screening of urine human serum albumin(HSA) is significant for the early diagnosis and intervention of the nephropathia.Surface Plasmon resonance(SPR) biosensor has the prospect to be applied in this field owing to high sensitivity,real time and label free working environment.This study developed sensitive HSA detection SPR biosensor assays.Different assay formats were investigated and compared.The calibration curves were constructed with the detection range above 0.1μg/mL for direct assay and between 1-50μg/mL for inhibition assay.The relative response could be amplified by using secondary antibody. |
