SDFl-3'A在云南11个少数民族群体中的分布

目的 为调查HIV-1感染相关等位基因SDF1-3'A在我国云南省11个少数民族群体中的频率和多态性分布.研究的人群包括599例中国少数民族群体个体(来自8个民族),他们均为健康无血缘关系个体.方法 从上述人群外周血中抽提基因组DNA,然后采用PCR和PCR-RFLP等方法进行基因分型.结果 411例是野生纯合子基因型,160例为杂合子基因型,28例为突变纯合子基因型.上述各群体等位基因型的分布符合Hardy-Weinberg平衡.11个民族群体的平均突变基因频率为0.176,等位基因频率范围分布在0.09～0.28之间,11个民族群体之间差异具有统计学意义(P<0.05).值得注意的是云南特有的独龙族突变基因频率最低为0.091.结论 单独从SDF1-3;A这一与艾滋病相关的遗传位点来分析,个体对于HIV病毒的抵抗保护性与目前中国人群艾滋病的流行病学状况并不一致.大部研究群体的突变基因频率均低于汉族群体,该突变基因在艾滋病发病过程中的影响值得进一步深入研究.

Distribution of SDF1-3'A Alleles in 11 Chinese Ethnic Populations of Yunnan Province

Objective To investigate the frequencies and polymorphisms of HIV-l resistant SDF1-3'A alleles in 11 Chinese populations in Yunnan Province.The blood samples include 599 unrelated healthy individuals.Methods Whole blood samples were collected randomly and their genomic DNA were extracted with 0MEGA Blood Kits.Allelic frequencies were identified by PCR.RFLP analysis.Results 411 individ. uals were wild-type homozygous.160 individuals were sDF-3'A heterozygote and 28 individuals were SDF1 3'A homozygous.Distributions of the SDF1-3'A alleles among the populations were in accordance with Hal-dy-Weinberg equilibrium.The average allele frequencies of total populations was 0.176,and the frequency span of the alleles was 0.09～O.28.Trhere were statistical differences among 11 populations P<0.05.A-mong them.the Drung ethnic population had the lowest SDF1-3'A allele frequency.Conclusion The pro-teetive effect of the SDF1-3'A allele in 11 populations has no accordance with the epidemic of AIDS in Yunnan Province.The frequencies of SDF1-3'A allele of 11 ethnic populations ale lower than that of Han.The mutant alleles conferring genetic resistance to HIV-1 infection and AIDS progression needs to be further studied.
