摘    要:间接荧光抗体技术(IFAT)已广泛用于肾综合征出血热(HFRS)病毒抗原的检测,其不足之处是需用荧光显微镜,有一定主观性及不能检测可溶性抗原。本文建立了以同样的抗HFRS病毒单克隆抗体(McAb)作为包被和第二抗体,以酶标葡萄球菌A蛋白(SpA)作指示抗体的双McAb ELISA间接夹心法,初步用于感染细胞培养上清及组织标本中HFRS病毒抗原的检测,获得较满意结果。

Abstract:A double McAb ELISA indirect sandwich method has been developed for detection of HFRS virus antigen. Dynamic changes of HFRS virus antigen in the cells infected and their culture supernatants were observed by IFAT and ELISA separatly. It was shown that HFRS virus antigen in the cells peaked 8 days after infection, whereas the antigen in the supernatants peaked 14 days after infec.tion. A hundred and seventy-nine brain and lung specimens of sucking mice artificially infected with HFRS virus were examined by ELISA and IFAT. The positive rates were 72.1% and 68.2%, respectively and the difference was significant ( P<0.05 ). The above results suggest that ELISA is a specific,sensitive and simple method, and can be used for detection of the viral antigen on the etiological and epidemiological studies of HFRS.
