不同齿科合金对成纤维细胞增殖凋亡的影响及机制研究

摘    要:目的 探讨不同齿科合金对成纤维细胞增殖凋亡的影响及机制。方法 小鼠成纤维L929细胞分为阴性对照组、金合金组、镍铬合金组和铜合金组,各组细胞培养48h后,MTT实验检测细胞增殖;流式细胞术检测细胞周期和细胞凋亡率;Western blot法检测Bcl-2、Bax、cleaved caspase-3蛋白表达。结果 金合金组和镍铬合金组细胞增殖率与阴性对照组比较,差异无统计学意义(P>0.05),铜合金组细胞细胞增殖率显著低于阴性对照组(P<0.01);金合金组G1期、S期和G2期细胞与阴性对照组差异无统计学意义(P>0.05),镍铬合金组和铜合金组G1期细胞显著低于阴性对照组,S期和G2期细胞显著高于阴性对照组(P<0.05);金合金组细胞凋亡率及Bcl-2、Bax、cleaved caspase-3蛋白表达与阴性对照组差异无统计学意义(P>0.05),镍铬合金组和铜合金组细胞凋亡率及Bax、cleaved caspase-3蛋白表达均显著高于阴性对照组,Bcl-2蛋白表达显著低于阴性对照组(P<0.01)。结论 金合金组、镍铬合金组和铜合金组对成纤维细胞增殖凋亡均有一定的影响,金合金组的影响最小,不同齿科合金引起成纤维细胞凋亡的机制可能与调控Bcl-2、Bax、cleaved caspase-3蛋白表达有关。

Abstract:Objective To investigate the effects and its mechanism of different dental alloys on proliferation and apoptosis of fibroblast. Methods Mouse fibroblast L929 cells were divided into negative control group, gold alloy group, Ni Cr alloy group and copper alloy group. Cell proliferation was detected by MTT assay. Cells were cultured for 48h. Cell cycle and apoptosis rate were detected by flow cytometry. The expression of Bcl-2, Bax, cleaved caspase-3 protein were detected by Western blot. Results Cell proliferation rate had no significant difference in gold alloy group and Ni Cr alloy group and negative control group (P>0.05), while was significantly lower in copper alloy group than the negative control group (P<0.01).G1 phase, S phase and G2 phase cells between gold alloy group and the negative control group had no significant difference (P>0.05), while G1 phase cells in Ni Cr alloy and copper alloy group was significantly lower than negative control group. S phase and G2 phase cells were significantly higher than the negative control group (P<0.05 or P<0.01). Cell apoptosis rate and the expression of Bcl-2, Bax, cleaved caspase-3 protein between gold alloy group and negative control group had no significant difference (P>0.05).Cell apoptosis rate and the expression of Bax,cleaved caspase-3 protein between Ni Cr alloy and copper alloy group were significantly higher than the negative control group. Bcl-2 protein expression was significantly lower than the negative control group (P<0.01). Conclusion The gold alloy, Ni Cr alloy and copper alloy can influence proliferation and apoptosis of fibroblast. The gold alloy group is minimum. Different dental alloys leads to the possible mechanism of fibroblast apoptosis was regulation of Bcl-2, Bax, cleaved caspase-3 protein expression.
