摘    要:目的应用~1H-NMR代谢组学技术比较线纹海马、大海马、三斑海马中化学成分的差异性,为中药海马质量评价研究提供依据。方法采用~1H-NMR代谢组学技术对3个不同品种海马药材进行NMR氢谱分析,归属、指认图谱中化合物,通过多元统计分析比较不同品种海马化学成分,发现其中的差异及其规律。结果在海马甲醇-水(1∶1)提取物~1H-NMR图谱中共指认出33种化学成分;多元统计分析结果显示线纹海马、大海马化学成分相近,差异性较小,而三斑海马相比于线纹海马、大海马,异亮氨酸、甲硫氨酸、谷氨酸、羟脯氨酸、脯氨酸、精氨酸、半胱氨酸含量降低,呈显著性差异,牛磺酸、甘油含量则显著上升。结论应用代谢组学技术从整体水平比较不同品种海马的化学成分,发现其中的差异性及其规律,为中药海马质量控制研究提供了新思路。

Abstract:Objective To compare the chemical constituents in Hippocampus kelloggi, H. kuda and H. trimaculatus by 1H-NMR metabonomics approach, provides the basis for quality evaluation of Hippocampus.Methods NMR-based metabonomics approach was used to analyze three breeds of Hippocampus and identify the chemicals. Multivariate statistical analysis was used to find the difference and regularity in the comparison of chemical constituents in different breeds of Hippocampus.Results Thirty-three metabolites were identified in the NMR spectra with methanol-ultrapure water (1:1); The multivariate analysis revealed that there were little differences between H. kelloggi and H. kuda; Compared with them, H. trimaculatus has the lower level of isoleucine, methionine, glutamic acid, hydroxyproline, proline, arginine, and cysteine, and the higher content of taurine and glycerin, which existed significant difference.Conclusion Compared the chemical constituents of different breeds of Hippocampus from the overall level to find their otherness and regularity, providing a new idea to control the quality of Hippocampus.
