黄芪注射液合川芎嗪注射液治疗类风湿关节炎的作用机理研究

目的观察黄芪注射液与盐酸川芎嗪注射液并用对佐剂性关节炎(AA)大鼠抗炎消肿的作用,探讨其机制。方法 70只健康Wistar大鼠随机分为正常组、模型组、黄芪组、川芎组、配伍组、甲氨蝶呤(TMX)组、雷公藤组。除正常组外,其余各组均建立AA大鼠模型,并于造模1周后分别给予相应的药物灌胃,每日1次,连续3周。观察大鼠一般情况,定期测量大鼠右后足肿胀度,检测大鼠血清中巨噬细胞移动抑制因子(MIF)、血管内皮生长因子(VEGF)、白介素-17(IL-17)、γ干扰素(INF-γ)含量的变化。结果模型组大鼠右后足趾关节肿胀明显,血清中促炎细胞因子MIF、IL-17、VEGF以及INF-γ的含量明显升高。与模型组相比,各给药组均可改善关节肿胀度,降低血清中炎性细胞因子MIF、IL-17、VEGF及INF-γ的含量,但配伍组疗效优于黄芪、川芎组,与雷公藤多苷片效果相当。结论黄芪注射液合川芎嗪注射液并用能控制AA大鼠病情的发展,其机制可能与对炎性细胞因子MIF、IL-17、VEGF及INF-γ的有效抑制有关。

Research of Astragalus Injection and Ligustrazine Injection in Action Mechanism of Rheumatoid Arthritis

Objective: To observe the effect of astragalus injection and ligustrazine hydrochloride injection and to investigate the intrinsic mechanism of the compound medicine on treatment of adjuvant-induced arthritis(RA).Methods:70 healthy wistar rats were randomly divided into the normal group,the model group,the astragalus root group,the ligustrazine injection group,the compatibility group,the MTX group and the TPT group.Besides the normal group,other groups were made the rat model of AA.After models establishment for a week,corresponding drug gavage were given in these groups one time a day for 3 consecutive weeks.There were some indicators were detected and compared such as the rats′ general situation and the regularly measure rat right rear foot swelling.These indices were detected inculding the content of cytokine MIF,IL-17,VEGF,INF-γ in rat serum.Results: The right rear toe joints of the rats in the model group were swelled significantly.The contents of MIF,IL-17,VEGF,INF-γ in the rats’ serums of the model group were increased significantly.Compared with the model group,all drug groups improved the right rear toe joints swelling degree and reduced the content of inflammatory cytokines MIF,IL-17,VEGF,INF-γin the rats′ serums.But the compatibility effect was better than astragalus root group and ligustrazine injection group.Conclusion: The effect of astragalus injection and ligustrazine injection could control the disease development of AA rats.The mechanism might be related to the inflammatory cytokines of MIF,IL-17,VEGF,INF-γ effective suppression.
