增生结节、肝硬变及肝癌组织中原癌基因 C-erbB-2 蛋白产物的表达

①目的了解原癌基因Ｃ－ｅｒｂＢ－２蛋白产物在肝细胞癌发生过程中的表达及其关系。②方法应用抗原癌基因Ｃ－ｅｒｂＢ－２蛋白产物的抗体，检测４１例人肝细胞癌（１８例有癌周组织）的抗原表达。③结果Ｃ－ｅｒｂＢ－２在肝细胞癌及癌周增生结节中的阳性表达率分别为２６．８３％（１１／４１）和２８．５７％（４／１４），两者间差异无显著性（χ２＝０．０２，Ｐ＞０．０５），但两种均高于肝硬变组织（χ２＝４．０２，Ｐ＜０．０５，直接概率计算，Ｐ＝０．０３）。癌周肝硬变组织Ｃ－ｅｒｂＢ－２表达阴性。④结论Ｃ－ｅｒｂＢ－２基因的活化与人肝癌发生有关，且Ｃ－ｅｒｂＢ－２过度表达是肝癌发生过程中的早期表现。同时也证实了增生结节属癌前期病变，与肝癌的发生有关；肝硬变与肝癌的发生没有直接关系。

EXPRESSION OF C erbB 2 ONCOGENE PROTEIN IN HYPERPLASTIC NODULES LIVER CIRRHOSIS AND HEPATOCELLULAR CARCINOMA

Objective To study the expression and significance of oncogene C erbB 2 protein in the development of hepatocellular carcinoma. Methods The expression of C erbB 2 oncogene protein was studied in 41 cases of human hepatocellular carcinoma(18 of them with surrounding tissues) by using specific antibody against the C erbB 2 protein. Results The positive rates of C erbB 2 oncogene protein expression in hepatocellular carcinoma and in the pericarcinomatous hyperplastic nodules were 26.83% (11/41 cases) and 28.57%(4/14 cases) respectively, there was no statistically significant difference between both of them. In 17 cases of liver cirrhosis in pericarcinomatous tissues, the expression of C erbB 2 oncogenes were negative. Conclusion Activated C erbB 2 oncogenes may have a role in human hepatocarcinogenesis and C erbB 2 overexpression is an early event in the development of hepatocellular carcinoma. Hyperplastic nodules are preneoplastic lesions and related to hepatocarcinogenesis; the liver cirrhosis does not correlate directly with hepatocarcinogenesis.
