Abstract:To our knowledge,this is the first study in China correlating polymerase chain reaction (PCR),virus isolation and DNA hybridization in cerebrospinal fluid (CSF) for diagnosis of herpes simplex virus encephalitis (HSE).7 CSF specimens from clinically definate HSE patients and 14 from the patients with other neurological diseases(OND) were involved. Herpes simplex virus (HSV) DNA was detected by PCR in 5 of 7 patients with viral encephalitis and in 3 of the 5 HSV DNA PCR positive patients,HSV-DNA was detectable by dot hybridization,while 2 strain of HSV were isolated from the 5 PCR positive samples.All the samples obtained from patients with OND showed nestive by the 3 techniques.The resulls suggested using PCR instead of virus isolation and DNA hybridazition might be a reliable and practical for early and rapid diagnosis of HSE because of its high sensitivity and specificity.
