微量板培养法在O139群霍乱疫情中的应用与比较

摘    要:目的:通过采用霍乱弧菌常规培养法与微量板培养法,对一起O139群霍乱疫情现场疫源标本进行比对检测研究,以探讨微板法替代常规法的可能性。方法:用霍乱弧菌常规培养法口与霍乱弧菌微量板培养法心,对某地O139群霍乱疫情现场采集的442份疫源检索样品,同时进行病原学检测,对比观察检测结果。结果:两种方法同时检测442份样品,共检出O139群霍乱弧菌18株,其中常规法检出16株,微板法多检出2株为18株。阳性检出率分别为3.62%和4.07%,阳性符合率为88.89%,检出率之间差别无统计学意义。结果显示微板法稍好于常规法,并且微板法还具有以下优点:1)灵敏度高;2)抗原凝集性好;3)培养板体积小、培养基量少(每孔仅1ml)、容量大(每板可做24份样品);4)操作简便,不需要特殊设备;5)微量板使用后,经消毒清洗可回收再利用,节约成本,在工作量大时,其实用、简便的优点更为突出。结论:微板法可以用于霍乱弧菌病原检测,可靠性较高,且检测霍乱的特异性和敏感性已达到卫生部《霍乱防治手册》的要求。

Abstract:Objective:To study on the probability to substitute the ordinary cultivation with the micro-plate cultivation through comparative use of the two methods in an outbreak of O139 cholera.Methods:To compare the ordinary cultivation and the micro-plate cultivation through detecting the pathogen among 442 samples collected from an O139 cholera outbreak.Results:Eighteen strains of O139 vibrio cholera were isolated from 442 samples,of which 16 strains were isolated by the ordinary method,18 by the micro-plate method with 3.62% and 4.07% of positive detection rate respectively.The positive conformity rate of the results was 88.89% with no statistically significance between the results.The micro-plate is of the following advantages: a: Higher sensitivity;b: A better antigenic agglutination;c: Less medium(less than 1 ml in a hole) with more sample detections ability(24 samples in a plate);d: Simple performance with no special equipment required;e: Economical: the micro-plate can be re-used after disinfection,much practical,particularly in a great amount of work.Conclusion:The micro-plate cultivation method is reliable for cholera pathogen detection with high specificity and sensitivity which have met the requirements of the "manual of cholera cure and prevention" edited by the Ministry of Health.
