5-LOX表达与胃癌淋巴结转移的关系及AA861对其表达的影响

目的探讨5-脂氧合酶（5-Lipoxygenase，5-LOX）在胃癌组织中的表达及其与肿瘤淋巴结转移的关系，以及5-LOX选择性抑制剂AA861对其表达的影响。方法采用RT-PCR方法检测30例胃癌标本及相匹配的癌旁组织中5-LOX mRNA的表达；用免疫组织化学方法检测了40例胃癌石蜡标本及30例癌旁组织石蜡标本中5-LOX蛋白的表达；免疫细胞技术和图象分析软件检测胃癌AGS细胞中5-LOX蛋白的表达水平及AA861对其表达的影响。结果胃癌组织中5-LOX蛋白和mRNA水平的表达均显著高于癌旁组织（P〈0．05）。伴淋巴结转移者5-LOX mRNA及蛋白表达高于无转移者（P〈0．05）。AA861干预组与对照组5-LOX蛋白表达无明显改变（P〉0．05）。结论5-LOX基因在胃癌中表达增高并与淋巴结转移明显相关，AA861对5-LOX蛋白表达无明显影响。

Correlation of 5-LOX expression to lymph node metastasis in human gastic cancer and the effect of AA861 on its expression

Objective To investigate the relationship between expression of 5_LOX mRNA,protein and lymph node metastasis of gastric cancer,and to observe the effect of AA861 on the expression of 5_LOX protein in human gastric carcinoma cell line AGS.Methods RT_PCR and immunohistochemical methods were used to examine the expression of 5_LOX mRNA and protein in gastric cancer and paracancerous tissue, respectively. The effect of AA861 on 5_LOX expression protein was studied with immuocytochemistry as well as images analyzing system.Results Expression of 5_LOX mRNA and protein in gastric cancer were significant higher than those in cancerous beside tissue(P<0.05).Expression of 5_LOX in gastric carcer with lymph node metastasis was significantly higer than that without metastasis(P<0.05). Expression of 5_LOX as no significant change between AA861 treated AGS and control group(P>0.05 ).Conclusion 5_LOX gene expression were increased in gastric cancer and associated with lymph node metastasis.AA861 had no effect on 5_LOX protein expression.
