组胺受体激动剂和拮抗剂对粒单系祖细胞增殖的影响

摘    要:用体外琼脂培养法研究了2-噻唑乙胺、dimaprit及甲氰咪胍对小鼠CFU—GM产率的影响。结果表明:10~(-8)M~10~(-4)M的2-噻唑乙胺不影响CFU—GM产率,而10~(-8)M的dimaprit即可明显增加CFU—GM产率。此作用随剂量的递增而加强,至10~(-5)M时,增至最大值,此后不再增高。单用甲氰咪胍不影响CFU—GM产率,但伍用dimaprit时,甲氰咪胍阻断dimaprit的上述作用;二者对CFU—GM产率的影响呈现竞争现象。分析了该结果的理论意义和临床意义。

EFFECTS OF THE AGONISTS AND ANTAGONIST OF HISTAMINE RECEPTORS ON THE PROLIFERATION OF CFU-GM

Abstract:The effects of 2-(2-Aminoethyl) Thiazole (2-AT), an agonist of histamine H1-receptor, dimaprit, an agonist of histamine H2-receptor, cimetidine, an antagonist of histamine H2-receptor, on the proliferation of granulocyte-macrophage progenitors (CFU-GM) of LACA mouse bone marrow were investigated by using the agar culture technique in vitro.It was found that 2-AT, at concentrations between 10-8M-10-4M, had no effect on the colony formation of CFU-GM.In contrast with that, dimaprit, at concentration of 10-8M, could significantly increase the colony numbers of CFU-GM as compared with the untreated controls (P < 0.05) .With the increase of the concentration of dimaprit, the colony numbers of CFU-GM increased steadily.The colony numbers reached the utmost limit when the concentration of dimaprit had gotten to 10-5M.Then, the colony numbers changed very little , although the concentration of dimaprit had increased further.It was also found that cimetidine alone had no significant effect on the colony formation of CFU-GM, but it could significantly block the effect of dimaprit on the production rate of CFU-GMwhen cimetidine had been used together with dimaprit. Dimaprit and cimetidine had competitive influence on the proliferation of CFU-GM. These results suggested that H2-receptors were present on the surface of CFU-GM.The stimulation of H2-receptors with their agonists enhanced the proliferation of CFU-GM, while the antagonist alone had no effect on the proliferation of CFU-GM, but the antagonist could block such effect of the agonist.H1-receptors alone may not influence the proliferation of CFU-GM.
