TFPI-2基因对胰腺癌Panc-1细胞增殖和侵袭能力的影响

摘    要:目的探讨外源性人组织因子途径抑制物(TFPI)-2基因表达对胰腺癌Panc-1细胞增殖和侵袭转移能力的影响。方法将人TFPI-2基因转染胰腺癌Panc-1细胞。用RT-PCR、Western blot技术检测目的基因的表达,用细胞生长试验、流式细胞仪分析技术以及Boyden小室法检测Panc-1细胞转染前后细胞增殖、细胞周期、细胞凋亡和细胞体外侵袭能力的变化。结果TFPI-2基因能在Panc-1细胞中稳定表达;与对照组相比,转染后的Panc-1细胞生长受到抑制,阻滞于G0-G1期,早期细胞凋亡率增加,细胞体外侵袭能力受到抑制。结论TFPI-2基因表达能够在体外抑制胰腺癌细胞系Panc-1的细胞增殖,降低其体外侵袭转移能力。

Abstract:[Objective] To evaluate the effect of TFP1-2 gene on cell proliferation and invasive ability of hmnan pancreatic cancer cell line Pane-1. [Methods]TFP1-2 gene was transfected into Pane- 1 cell, TFPI-2 mRNA and protein expression were detected by RT - PCR and Western blot. MTT assay was used to assess the effect of TFPI - 2 on cell proliferation. The change of cell cycle and apoptosis were measured by flow cytometry and the invasive ability in vitro was detected by Boyden chamber. [ Results ] RT - PCR and Western blot analysis showed that Panc - 1 cell could express TFPI - 2 stably. Compared with non - transfected cells, the growth of Panc - 1 cell transfected with TFPI - 2 was inhibited significantly. The transfected cells showed a significant increase in GO - G1 phase and could be induced to apoptosis of in comparison with the control group. The invasive ability of Panc - 1 cell was inhibited by expression of TFP1-2 effectively. [Conclusions] The expression of TFPI - 2 can inhibit cell proliferation and invasive ability of human pancreatic cancer cell line Panc - 1.
