半边莲生物碱对肺动脉高压大鼠ET-1信号通路的影响

摘    要:目的 观察半边莲生物碱对肺动脉高压(PAH)大鼠肺小动脉重构的影响并探讨其机制.方法 24只雄性Wistar大鼠随机分为正常对照组、模型组和半边莲生物碱组,每组8只.腹腔注射野百合碱(MCT)复制PAH模型.半边莲生物碱干预4周后, HE染色观察肺小动脉病理学变化,ELISA法检测大鼠血浆内皮素-1(ET-1)水平,免疫组化法检测肺组织内皮素受体A(ETA)的表达.结果 半边莲生物碱可显著改善肺小动脉重构,降低血浆ET-1水平及肺组织ETA的表达.结论 半边莲生物碱可抑制MCT所致的PAH大鼠肺小动脉重构,这可能与其显著降低大鼠血浆ET-1水平及肺组织ETA的表达有关.

Abstract:Objective To observe the effect of Lobelia alkaloids on pulmonary vascular remodeling of pulmonary arterial hypertension (PAH) rat model and to explore its mechanism. Methods Twenty-four male Wistar rats were randomly divided into normal control group, model group and Lobelia alkaloids group. PAH rat models were established by giving monocrotaline (MCT) intraperitoneally, and the rats were given Lobelia alkaloids by intragastric administration for 4 weeks. HE staining, ELISA and immunohistochemical methods were used to study pulmonary morphological changes, endothelin-1 (ET-1) level in plasma and endothelin receptor A (ETA) expression in lung tissue. Results Lobelia alkaloids could significantly alleviate pulmonary artery remodeling and decrease the ET-1 level in plasma and ETA protein expression in lung tissue of PAH rat model. Conclusion Lobelia alkaloids can inhibit significantly pulmonary artery remodeling of PAH rat model induced by MCT which may be related to the decreased ET-1 level in plasma and ETA protein expression in lung tissue.
