摘    要:目的 探讨酒精对锥体外系影响机制。方法 48只8周龄雌性幼鼠随机分成对照组和低、中、高浓度组,分别用不同浓度的酒精(0%、12.5%、25%和50%)灌喂,按10 ml/kg,隔天1次,90 d后,分别对各浓度组小鼠进行灌酒前、后转棒试验。转棒试验结束后快速取出小脑和尾壳核,并测量它们的乙酰胆碱酯酶(AchE)活性。结果 高浓度组小鼠灌酒前、后在棒上停留时间均减少,中浓度组小鼠灌酒后在棒上停留时间也减少;高、中浓度组小脑和尾壳核胆碱酯酶活性降低。结论 酒精致锥体外系功能障碍与小脑、尾壳核的胆碱酯酶活性降低有关。

Abstract:Objective To explore the mechanism of ethanol exposure on the extrapyramidal system of mouse. Methods Forty-eight mouse(8W) were divided randomly into the control group, the low middle and high dose groups. Different dose ethanol(0%, 12.5% and 50% ) were given(10 ml/kg) intragastrically by gavage once every other day for 90 days respectively. The rotarods were tested before and after ethanol administration. Mouse were sacrificed by decapitation after the rotarod test. Cerebellum and caudate putamen were rapidly removed. Acetylchohnesterase activity in cerebellum and caudate putamen was assayed. Results Both before and after exposure to ethanol the sustaining time of rotarod of the high dose group were shorter, and that of the middle dose group after exposure to ethanol was also shorter; acetylcholinesterase activity of the cerebellum and caudate putamen in the high, middle group decreased. Conclusion There is the relation between the malfunctions of extrapyramidal system caused by ethanol and the decrease of acetylcholinesterase(AchE) activity in cerebellum and caudate putamen.
