电针对慢性应激模型大鼠下丘脑及胃肠道NMDA受体亚单位mRNA表达的调控作用

摘    要:目的:从NMDA受体亚单位NR1、NR2B的角度,探讨慢性应激状态下大鼠下丘脑、胃肠道受体后信号转导的变化及电针百会、印堂、天枢穴改善抑郁症消化功能障碍的作用机理。方法:将40只雄性SD大鼠随机分为空白组、模型组、对照组、电针组,每组10只。采用RT—PCR实验方法分析慢性应激模型大鼠下丘脑、胃窦及空肠组织NMDA受体亚单位NR1、NR2BmRNA表达的变化,及电针印堂、百会和天枢穴的干预作用。结果:模型组大鼠下丘脑、胃窦及空肠组织NR1、NR2BmRNA表达与正常组相比明显升高(P〈0.05),经治疗后,电针组下丘脑、胃窦及空肠组织表达明显下降(P〈0.05)。结论:抑郁模型大鼠下丘脑、胃窦及空肠组织NMDA受体亚单位NR1、NR2BmRNA表达上调,电针改善抑郁症伴发的消化功能障碍可能与调节NMDA通路有关。

Abstract:Objective:To observe the changes of signal transduction and the effect of electro - acupuncture (EA) at Baihui, Yintang, Tianshu points on the rat models with depression from the point of view of the content and activity of NR1Mrna and NR2BmRNA. Methods:Forty male Sprague Dawley rats were randomly divided into four groups: control group, depression model group, western medicine group and EA group, with ten rats in each group. Observe the image of NRlmRNA, NR2BmRNA in hypothalamus, gastric antrum and ileum in the rats with depression and the effect of EA by the technique RT - PCR. Results: The image of NRlmRNA, NR2BmRNA in hypothalamus, gastric antrum and ileum of the rat models were higher than that in normal group (P 〈 0.05 ). EA could markedly decrease them after treatment(P 〈 0.05 ). Conclusion:The signal transduction of NR1Mrna and NR2BmRNA is higher in hypothalamus, gastric antrum and ileum in the models with depres- sion. The modulation to the functional dyspepsia caused by depression of EA may be related to its function of adjusting the signal transduction of NMDA.
