还少丹对D-半乳糖衰老模型小鼠白介素-2及线粒体DNA含量的影响

摘    要:目的:探讨还少丹对衰老模型小鼠白介素-2(IL-2)及线粒体DNA(mitochondrial DNA,mtDNA)含量的影响。方法:将40只小鼠随机分成空白对照组、衰老模型组、低剂量药物组和高剂量药物组,每组10只。采用皮下注射D-半乳糖溶液(100 mg/kg)制备亚急性衰老模型,同时用还少丹水煎液灌胃6周。双抗夹心ELISA法测定血清IL-2的含量;分光光度法测定肝、脑组织中mtDNA含量。结果:与空白对照组比较,模型组小鼠血清IL-2含量明显减少,肝和脑组织mtDNA含量明显增高(均P<0.01)。与模型组比较,还少丹低剂量和高剂量组都能提高衰老模型小鼠血清IL-2的含量,降低肝、脑组织mtDNA的含量,差异均具有统计学意义(均P<0.01)。结论:还少丹能够提高衰老小鼠的机体免疫功能,降低肝、脑组织中mtDNA的含量,从而起到延缓衰老的作用。

Abstract:Objective: To explore the effect of Huanshaodan on the content of interleukin-2(IL-2) and mitochondrial DNA(mtDNA) in aging model mice.Methods: Forty mice were randomly divided into 4 groups:a blank control group,a aging model group,a low dose Huanshaodan group and a high dose Huanshaodan group(all n=10).The aging model mice were established by D-galactose(100 mg/kg) hypodermic injection for 6 weeks.Huanshaodan had been administered in the low dose and high dose Huanshaodan groups.Then serum levels of IL-2 was detected by a double-antibody sandwich enzyme-linked immunosorbent assay,while mtDNA content of liver and brain was delected by spectrophotometry.Results: Compared with the blank control group,serum levels of IL-2 significantly decreased in the aging model group,and the mtDNA content of liver and brain were significantly increased(all P<0.01).Compared with the aging model group,the serum levels of IL-2 obviously increased,and mtDNA content of liver and brain significantly decreased in both the low dose and high dose Huanshaodan groups(all P<0.01).Conclusion: Huanshaodan can improve the immune function of aging mice,decrease the mtDNA content of liver and brain,and thus play a role in delaying aging.
