桦褐孔菌提取物对人胃癌BGC-823细胞株的抑制增殖及诱导凋亡作用

摘    要:目的]观察桦褐孔菌提取物对人胃癌BGC-823细胞株的抑制增殖及诱导凋亡作用.方法]选用人胃癌细胞株BGC-823进行体外培养,用MTT法测定桦褐孔菌提取物对BGC-823细胞株的增殖抑制率,应用倒置显微镜、HE染色光学显微镜、AO/EB双染色荧光显微镜及透射电子显微镜等仪器观察桦褐孔菌提取物对肿瘤细胞形态结构的影响.结果]10,20,40,80 mg/L桦褐孔菌提取物对胃癌BGC-823细胞株有抑制增殖作用,且表现出浓度依赖关系.HE染色在光学显微镜下可见桦褐孔菌组肿瘤细胞体积变小、变圆,核染色质浓缩或形成块,部分细胞膜形成凋亡小体;在倒置显微镜下可见桦褐孔菌组肿瘤细胞体积变小、变圆,核染色质凝集,细胞间连接疏松,贴壁能力明显减弱;在AO/EB双染色荧光显微镜下可见桦褐孔菌组肿瘤细胞胞核染色质呈橘红色,胞质内的橘红色或橘黄色荧光消失,细胞周围有呈亮绿色的荧光凋亡小体;在透射电子显微镜下可见80 mg/L桦褐孔菌提取物组肿瘤细胞核染色质固缩边集,胞质内可见有空泡,凋亡小体凸出于细胞表面.结论]桦褐孔菌提取物可抑制人胃癌BGC-823细胞株的增殖,诱导肿瘤细胞凋亡.

Abstract:OBJECTIVETo observe the effects of cell anti-hyperplasis and apoptosis of BGC-823 human gastric carcinoma cells in vitro after treatment with extracts from Inonotus obliquus.METHODSBGC-823 human gastric carcinoma cells were cultured in vitro.MTT assay was used to determine the effect of extracts from Inonotus obliquus on cell growth inhibitory.Morphological changes of the apoptosis of BGC-823 cells were observed by HE staining,AO/EB double fluorescent staining through invert microscope,light microscope,fluorescence microscope,and ultrastructure of BGC-823 cells were observed under transmission electron microscope.RESULTSLTS extracts from Inonotus obliquus in concentrations of 10,20,40,80mg/L could inhibite the hyperplasis of BGC-823 human gastric carcinoma cells in dose-dependent manner.The BGC-823 human gastric carcinoma cells treated with extracts from Inonotus obliquus by HE staining showed morphological changes such as cellular volume small and round,nuclear chromatin condensation,plasma membrane bleb formation,also their apoptosis body formations were observed under light microscope.The BGC823 human gastric carcinoma cells treated with extracts from Inonotus obliquus showed cellular volume small and round,nuclear chromatin condensation,loose between the cells and adherent ability obviously decrease under invert microscope.BGC-823 human gastric carcinoma cells were stained by AO/EB double fluorescent staining were observed under fluorescence microscope,appearing as chromatin orange-red condensation in nuclear,orange-red or orangeyellow fluorescent disappeared and light green fluorescent apoptotic bodies around cells after induction of extracts from Inonotus obliquus.After treatment with 80mg/L extracts from Inonotus obliquus,apoptosis of BGC-823 human gastric carcinoma cells were observed under transmission electron microscope, the nuclear chromatin was condensed or aggregated under the nuclear membrane,the membrane blebs appeared and apoptotic body formations bulged cells surface.CONCLUSIONExtracts from Inonotus obliquus exerts has the effects of anti-hyperplasis and apotptsis on BGC-823 human gastric carcinoma cells.
