摘    要:目的:研究大块牛松质骨(MBCB)处理后的各种成分及高浓度H2O2消除异种骨抗原性机制,方法:采用干燥失重法及殖渣法检测经过系统去抗原处理后的大块异常骨的有机,无机质及水份含量,将25块大块新鲜牛松质骨均分为5组,分别用高浓度H2O2浸泡0,24,48,72和96h,采用氨基酸分析方法研究不同H2O2处理时间的骨块中羟脯氨酸,脯氨酸含量,结果:经处理后,MBCB为色白,并呈现出多孔样结构,扫描电镜观察,松质骨呈清晰的多孔结构,孔壁光滑,5组检测样品中羟脯氨酸及脯氨酸的含量随有H2O2处理时间的延长均有同步下降的趋势,但各组间无显性差异(P>0.05),5组检测样品中18例氨基酸含量及氨基酸总量随着H2O2处理时间的延长亦有同步下降的趋势,仅0h与96h组间18种氨基酸以及总氨基酸含量有显性差异(P<0.05),其余各组间无显性差异(P>0.05),5组检测样品中羟脯氨酸及脯氨酸在总氨基酸总量所占比例随差H2O2处理时间的延长无明显变化,各组间无显性差异(P>0.05),结论:大块异种骨经系统去抗原处理后,保持了一定的有机及无机质比列,即具有良好的力学特性又消除了其抗原性,提示高氧化剂引起蛋白质变性是消除大块异种骨抗原性的主要原因。

Abstract:AIM To analyse the components of antigen-eliminated massive bovine cancellous bone (MBCB) and to study the mechanism of high concn H2O2 eliminating the antigenicity of xenogenic bone. METHODS Dry-weight lost method was applied to measure the organic, mineral, and water components in MBCB; 25 massive flesh bovine cancellous bone blocks were distributed into 5 groups, which were dipped in 300 mL*L-1 concn H2O2 for 0, 24, 48, 72, 96 h.The quantities of hydroxyproline and proline in the five groups were measured by amino acid analysis. RESULTS After the antigen eliminating program, MBCB occurred as white colour and porous structure. SEM showed the wall of bone trabecula in the center part of MBCB was quite clear. The quantities of hydroxyproline and proline decreased a little with the time treated in high concn H2O2 in the five groups, but no differences between the five groups were found; the same decreasing tendency was found to the quantities of 18 amino acids of the bone blocks in the five groups; difference was only found between 0 h and 96 h group (P<0.05). The percent of hydroxyproline and proline in amino acids was not found any difference in the five groups (P>0.05). CONCLUSION The MBCB may contain reasonable rate of organic and mineral material, which is beneficial to keeping the mechanic property of MBCB after its antigen-eliminating program. Denaturalization of protein in the condition of oxidation of high concn H2O2 would be the major mechanism to eliminate the antigenicity of massive xenogenic bone.
