Rg_2与R_f对钙通道作用的单通道分析 |

摘 要: | 在单个Wistar大鼠乳鼠心室肌细胞上,用连细胞电压钳法,观察人参三醇组皂甙单体Rg2、Rf60μmol·L-1对钙通道单通道活动的影响,并与钙通道阻滞剂异博定79μmol·L-1、钙通道激动剂BAK86445μmol·L-1对照,证明Rg2对钙通道有阻滞作用,其机理在于使钙通道的开放时间缩短、开放概率减少、关闭时间延长,而对Baa+流幅值无明显影响,Rf可缩短L型钙通道的开放时间、延长关闭时间、降低开放概率。

Abstract: | The influence of
panaxatrio1 saponin monomers Rg2、Rf 60 μmol·L-1 on single Calci-um channel activity
from cultured neonatal Wistar rat myocardiocyte was observed using the cell-at-tached
configuration of patch clamp technique,and compared with that of calcium channel blocker
ver-apamil 79 μmol·L-1 and calcium channel activator BAY K 8644 5μmol·L-1. The
inhibiting effectsof Rg2 on the three-type calcium channels were observed. The mechanism
was that Rg2 shortented theopen time ,decreased the open一state probability, lengthened the
close time of calcium channels,with-out influencing the amplitude of the ionic current flowing
through the opening calcium channel;Rf hadno these actions. |
