大黄碳酸氢钠片总蒽醌含量测定及指纹图谱研究

摘    要:目的对大黄碳酸氢钠片中5种蒽醌类成分进行含量测定,建立其HPLC指纹图谱。方法采用DIKMA Diamonsil C18(250 mm×4.6 mm,5μm)色谱柱;流动相为甲醇-1 mL·L^-1磷酸溶液(85∶15);流速:1.0 mL·min^-1;检测波长:254 nm。利用中药色谱指纹图谱相似度评价系统软件(2004A版)对大黄碳酸氢钠片HPLC图谱进行分析。结果3个厂家的16批大黄碳酸氢钠片样品中5种蒽醌类化合物含量有显著差异。16批样品指纹图谱相似度均在0.990以上,指认共有峰9个。结论该方法简便、准确、重复性好,可为大黄碳酸氢钠片的质量控制和评价提供科学依据。

Abstract:Objective To determine the contents of 5 anthraquinones in Rhubarb Sodium Bicarbonate Tablets and establish HPLC fingerprint.Methods The separation was performed on a DIKMA Diamonsil C 18(250 mm×4.6 mm,5μm)column.The mobile phase was methanol-1 mL·L^-1 phosphate solution(85∶15),the flow rate was 1.0 mL·min^-1,and the detection wavelength was 254 nm.Similarity evaluation system for chromatographic fingerprint of TCM(2004A)was used for establishing HPLC fingerprint and similarity evaluation.Results The contents of 5 anthraquinones in 16 batches of Rhubarb Sodium Bicarbonate Tablets samples from 3 manufacturers were significantly different.The similarity of fingerprints of 16 batches of samples were all above 0.990,and 9 common peaks were identified.Conclusion This method is simple,accurate and repeatable,which can provide scientific basis for quality control and evaluation of Rhubarb Sodium Bicarbonate Tablets.
