摘 要: | 2007年德国Merck公司宣布艾滋病病毒(HIV)疫苗失败后,艾滋病疫苗研究陷入低谷。2011年在泰国进行的RV144疫苗显示受试人群获得了31.2%的保护,这个出人意料的结果让研究者重燃信心,即有效的HIV疫苗是可能的。随后,抗HIV-1中和抗体产生条件、靶点、抗体结构及运用等,成为了近年的研究热点。

Abstract: | Antibodies that neutralize diverse strains of HIV 1 should be broad, potent and with long half- life. Research and development of new HIV-neutralizing monoclonal antibodies (mAbs) have been accelerated by new techniques, particularly the structure biology-guided antigen design with the deep understanding of the epitope structure on the HIV-1 envelope. To study the mechanism of inhibition, the specific features and epitope locations of the new generation mAbs is critical for improving the treatment, protection against HIV-1 infection and developing novel strategies for vaccine design. In this review, we focus on several HIV-neutrali- zing mAbs about their efficacy and potential strategies that will lead to more effective approaches against HIV-1. |
