重组血管内皮生长因子_(165)腺相关病毒对心肌梗死大鼠心功能的影响

摘    要:目的探讨直接心肌注射基因重组内皮生长因子_(165)腺相关病毒(rAAV-hVEGF_(165))对急性心肌梗死大鼠心功能的影响。方法选择雄性SD大鼠81只,将0.1 ml生理盐水或rAAV-hVEGF_(165)分3点注射于梗死交界处心肌内。根据注射药物的不同随机分为4组:假手术组(15只),心肌梗死组(25只),生理盐水组(25只),血管内皮生长因子(VEGF)组(16只)。注射4周后测定心肌组织VEGF含量、超声心动图参数、梗死面积、微血管数量、心钠素水平。结果 VEGF组大鼠心肌梗死面积较心肌梗死组和生理盐水组明显减小(P<0.05),左心室射血分数明显高于心肌梗死组和生理盐水组(P<0.01),心肌组织VEGF含量较心肌梗死组和生理盐水组有所增加。血管计数显示,VEGF组大鼠较其他3组有更多的新生血管形成(P<0.01)。结论直接心肌注射rAAV-hVEGF_(165)能够明显改善急性心肌梗死大鼠的心功能,缩小梗死面积,促进心肌内新血管的形成。

Effect of intramyocardial injection of rAAV-hVEGF165 on cardiac function of rats with myocardial infarction
Abstract:Objective To determine the effects of intramyocardial injection of rAAV-hVEGF_(165) on cardiac function in a rat model of acute myocardial infarction(AMI). Methods Eighty-one male SD rats were included. 0.1 ml of rAAV-hVEGF_(165) or saline was intramyocardially injected into three different sites at the border zone of infarction. The models were divided randomly into four groups: group sham operation (n = 15) ;group NS (n = 25) ; group MI (n = 25) ;group VEGF (n = 16). Echocardiogram data, infarct size, intramyocardial microvessels, VEGF in myocardium were measured 4 weeks after injection. Results Infarct size in group VEGF was significantly smaller than that in group MI and group NS (P<0.01). Echocardiogram data revealed that improvement of ejection fraction(LVEF%) was greater in group VEGF than in group MI and group NS (P<0.01). Vessel count showed that there were more intramyocardial vessels in group VEGF than in other 3 groups (P<0.01). Conclusions The research shows that intramyocardial injection of rAAV-hVEGF_(165) is capable of significantly improving cardiac function, decreasing infarct size,promoting myocardial neovascularization in a rat model of acute myocardial infarction.
