阿霉素对大鼠肺缺血再灌注损伤中血红素加氧酶-1的影响

摘    要:目的观察阿霉素对大鼠肺缺血再灌注损伤(LIRI)中血红素加氧酶-1(HO-1)的影响。方法32只雄性Wistar大鼠随机分成4组:手术对照组(C)、阿霉素组(ADM)、缺血再灌注组(IR)、阿霉素+缺血再灌注组(ADM-IR)。复制大鼠单侧肺缺血再灌注损伤模型,观察肺组织病理形态变化,对比观察各组肺干湿质量比(D/W)、肺泡损伤数(IAR)、HO-1活性、HO-1蛋白的表达。结果IR组与ADM-IR组D/W、IAR均比C组与ADM组高(P<0.01),但ADM-IR组显著低于IR组(P<0.01),C组与ADM组之间差异无统计学意义(P>0.05);与C组相比,ADM组、IR组与ADM-IR组HO-1活性、HO-1蛋白表达增加(P<0.01),ADM-IR组上升更加显著(P<0.01)。结论阿霉素能增加HO-1的活性,上调HO-1蛋白的表达,可能通过HO-1的抗氧化作用减轻肺缺血再灌注损伤;小剂量阿霉素对大鼠肺脏未造成明显损害。

Effect of adriamycin on heme oxygenase-1 in rat lung ischemia -reperfusion injury

Abstract:Objective To investigate the effect of adriamycin on heme oxygenase-1 in rat lung ischemia -reperfusion injury. Methods Thirty-two healthy male Wistar rats were divided randomly into 4 groups (eight rats in each group), namely the surgical controls (group C), Adriamycin group (group ADM), ischemia-reperfusion group (group IR) and ADM+IR group (group ADM-IR). The unilateral lung ischemia-reperfusion model was replicated. Rats from each group were sacrificed after a process of lung cross-clamping for 45 minutes followed by reperfusion for 2 hours, and were then harvested for the right lung for histological study. The dry/wet weight ratio of lung tissue (D/W), injured alveolus rate (IAR), the activity of HO-1, the expression of HO-1 protein were measured respectively in different groups. Results The value of D/W and IAR were much higher in IR group and ADM-IR group than those in C group and ADM group (P〈0.01). The values of these parameters were significantly lower in ADM-IR group than in the IR group (P〈0.01). There was no significant difference between ADM group and normal group (P〉0.05). The activity of HO-1 and expression of HO-1 protein were much higher in ADM group, IR group and ADM -IR group than those in C group (P〈 0.01), and these indexes in ADM-IR group increased more significantly (P〈O.01). Conclusion Adriamycin appeared to improve the activity of HO-1, up-regulate the expression of HO-1 protein. Ameliorate of lung IRI in rats may result from activation of HO-1 in lung tissue. Low-dose Adriamycin did not seem to cause evidenced injury in rat lung tissue.
