塞来昔布对N-丁基-N-(4-羟丁基)亚硝基胺诱导的大鼠膀胱癌抑制作用的研究

摘    要:目的探讨环氧化酶-2的抑制剂塞来昔布(celecoxib)对N-丁基-N-(4-羟丁基)亚硝基胺(BBN)所诱导的SD大鼠膀胱癌是否有抑制作用。方法本实验将SD大鼠随机分成3组,3组均以BBN为致癌剂诱发SD大鼠膀胱肿瘤的生长,同时,其中的两个实验组另外喂以不同剂量的celecoxib(500mgcelecoxib/kg饮用水、1000mgcelecoxib/kg饮用水)。30周后处死动物,观察celecoxib对BBN所诱导的膀胱癌是否有抑制作用以及不同剂量的celecoxib对膀胱癌形成的抑制作用是否不同。结果celecoxib能明显抑制BBN所诱导的SD大鼠膀胱肿瘤的形成(P<0.05)。剂量相对大的celecoxib对BBN所诱导的SD大鼠膀胱癌具有更强的抑制作用(P<0.05)。结论celecoxib对BBN所诱导的SD大鼠膀胱癌有抑制作用。

Abstract:Objective To detect whether cyclooxygenase-2 inhibitor celecoxib could prevent the SD mice urinary bladder cancer induced by N-butyl-N-(4-hydroxybutyl)-nitrosamine (BBN) or not. Methods SD mice were random divided into 3 groups. All the 3 groups were treated by BBN to induce urinary bladder cancer; meanwhile 2 of the 3 groups were fed by celecoxib in addition at different dosage (500mg celecoxib/kg tap water, 1000mg celecoxib/kg tap water). The animals were killed after 30 weeks to observe whether celecoxib can prevent the SD mice urinary bladder cancer induced by BBN or not and whether different dosage celecoxib can exert different effects or not. Results The celecoxib can obviously prevent the SD mice urinary bladder tumor induced by BBN (P<0.05). The high dosage celecoxib exerted more potent preventive effect against bladder cancer (P<0.05). Conclusion The celecoxib can prevent SD mice urinary bladder cancer induced by BBN.
