姜黄素含药血清对大鼠肝星状细胞HSC—T6增殖的影响

摘    要:目的观察姜黄素药物血清对瘦素刺激肝星状细胞增殖的影响,探讨其抗肝纤维化的机制。方法用姜黄素煎荆给大鼠灌胃(连续7d),制备含药血清,并用培养基将药物血清配制成四个稀释倍数,分别是5%、10%、20%、加%四个稀释倍数浓度;培养肝星状细胞,取对数生长期细胞,运用噻唑蓝(MTr)比色法检测药物血清对HSC—T6增殖的影响。结景姜黄素药物血清对增殖的肝星状细胞有抑制作用(P〈0.05或P〈0.01),在5%~40%的血清稀释倍数范围内,随剂量增大而抑制作用增强;在20%~40%的血清稀释倍数范围内姜黄素组对HSC—T6抑制率增高(P〈0.05)。结论姜黄素具有抑制HSC—T6增殖的作用。

Abstract:Objective to observe the effect of curcumin drag serum on proliferation of hepatic stellate cells stimulated by leptin, and investigate the mechanism of anti hepatic fibrosis. Methods using curcumin decoction to rats by gavage (7D), the preparation of serum containing medium, and drug serum was formulated in four dilution, respectively is 5 %, 10 %, 20 %, 40 % and four dilution concentration; hepatic stellate cells in culture, cells in the logarithmic growth phase, use of thiazide tetrazolium (MTI') ratio affects the detection drug serum on the proliferation of HSC - T6 colorimet- fie method. Results cureumin drug serum on proliferation of hepatic stellate cell inhibition ( P 〈0.05 orP 〈0.01 ), in 5 %- 40 % serum dilution range, while the inhibition enhanced with the increase of dose; high increases in the 20% - 40% of the serum dilution range of curcumin group on the inhibition rate of HSC - T6 ( P 〈 0.05). Conclusion curcu min can inhibit the proliferation of HSC - T6.
