蓄血证大鼠模型血清及血浆中炎症及凝血指标的研究

摘    要:目的研究蓄血证大鼠模型的炎症相关指标(IL-1β、IL-6、IL-4、IL-10、TNF-α、IFN-γ、SOD、MDA)及凝血相关指标(PT、TT、6-Keto-PGF1α、TXB2、ET-1、NO、t-PA、PAI-1)的变化,探讨相关指标与蓄血证的相关性。方法采用间隔24 h两次鼠尾注射内毒素脂多糖(LPS)复制蓄血证大鼠模型,观察大鼠体温、全血及血浆黏度变化,并检测相关炎症指标和凝血指标。结果在第二次给予LPS后,多数大鼠体温上升;2 h后大鼠的血液黏度增加最为显著;3 h内,模型组大鼠的PT、TT值先逐渐下降,然后又逐渐上升,其在2 h附近达到最低点。与正常组比较,模型组的2 h组、3 h组的血浆黏度、全血黏度差异有统计学意义(P〈0.01);模型组大鼠血清中TNF-α、IL-1、IL-6、IFN-γ值差异无统计学意义(P〉0.05),IL-10值、SOD值、MDA值、t-PA含量值、PAI-1值、t-PA/PAI-1比值差异均有统计学意义(P〈0.05,P〈0.01);模型组大鼠血浆中NO、6-Keto-PGF1α值、TXB2、6-Keto-PGF1α/TXB2差异均有统计学意义(P〈0.05,P〈0.01)),ET-1值差异无统计学意义(P〉0.05)。结论造模后2 h,大鼠血清及血浆中炎症及凝血相关指标的相应改变,表明大鼠呈现出"瘀""热"互结状态,与中医蓄血证有一致性。

Abstract:Objective To research the changes of inflammation indicators and coagulation indicators and explore the correlation between blood accumulation syndrome and the relative indicators.Methods LPS were injected into rats to duplicate rat models at every 24 hours.The temperature,blood viscosity and plasma viscosity were observed and the inflammation indicators and coagulation indicators were detected.Results Rats body temperatures were raised after LPS were injected at 2nd time;blood viscosity was increased markedly after two hours;within three hours,the PT value and TT value of the model group were decreased then increased that reached the lowest point in two hour.Compared with normal group,there were significant difference in the blood plasma viscosity and blood viscosity in 2 h model group and 3 h model group(P0.01);no significant difference was found in TNF-α,IL-1,IL-6,IFN-γ in the serum of the model group(P0.05),there were significant differences in IL-10,SOD,MDA,t-PA,PAI-1,t-PA/ PAI-1(P0.05,P0.01);there were significant differences in NO,6-Keto-PGF1α,TXB2,6-Keto-PGF1α/TXB2 in the serum of the model group(P0.05,P0.01),and no significant difference was found in ET-1 value(P0.05).Conclusion The inflammation and coagulation indicators were changed at 2 hours after the establishment of rat models.The coagulation of blood stasis and heat was found in rats,which in accord with the blood accumulation syndrome.
