核因子κB抑制因子基因多态性与非小细胞肺癌的关系

摘    要:目的探讨核因子κB抑制因子(I-κB)HaeⅢ基因多态性与非小细胞肺癌(NSCLC)的关系。方法选取110例NSCLC患者和110例年龄、性别相匹配的健康对照者,采用聚合酶链反应—限制性酶切长度多态性(PCR-RFLP)分析方法检测I-κB的HaeⅢ基因多态性分布,分析HaeⅢ基因多态性与罹患NSCLC癌危险性的关系。结果NSCLC组和正常对照组I-κB基因型分布符合Hardy-Weinberg平衡,两组的HaeⅢ基因型分布比较,差异有统计学意义(P〈0.05);G等位基因频率NSCLC组为61.8%,对照组为47.7%;A等位基因频率NSCLC组为38.2%,对照组为52.3%,差异也有统计学意义(P〈0.05);等位基因频率与NSCLC相对风险度分析显示G等位基因者患NSCLC是携带A等位基因者的1.773倍(OR=1.773,95%CI=1.225~2.474)。结论I-κB基因的HaeⅢ基因多态性影响罹患NSCLC的危险性,G等位基因是NSCLC的危险因素,携带等位基因G可能比携带等位基因A的人群更容易患病。

Abstract:Objective To investigate the associations of the Hae Ⅲ gene polymorphisms of IkappaB gene non-small-cell lung cancinoma(NSCLC).Methods The genotype of IkappaB was determined by PCR-restriction fragment length polymorphism in 110 NSCLC patients and 110 healthy control subjects that age and sex matched.To check the associations of the Hae Ⅲ gene polymorphisms of IkappaB gene and NSCLC risk.Results The genotype distribution of IkappaB gene in two groups wab consistent with Hardy-Weinberg balance.Significant difference was showed in the Hae Ⅲ genotype distribution between NSCLC patients and controls(P〈0.05).The frequencies of the G alleles of IkappaB gene were 61.8% in NSCLC group and 47.7% in control group.The frequencies of the A alleles of IkappaB gene were 38.2% in NSCLC group and 52.3% in control group,significant difference was also showed between A and G alleles frequency(P〈0.05).Risk for getting NSCLC in people carriers with G allele wab 1.773 times than people with A allele(OR=1.773,95%CI= 1.225~2.474).Conclusions The Hae Ⅲ gene polymorphism of IkappaB gene is associated with NSCLC risk,"G" allele may be a risk factor for NSCLC,people carriers with G allele easily get NSCLC than people with A allele.
