依达拉奉对老年痴呆大鼠学习记忆及海马4-羟基壬烯醛表达的影响

摘    要:目的:探讨依达拉奉对老年痴呆大鼠学习记忆及海马组织丙二醛(malondialdehyde,MDA)及4-羟基壬烯醛(4-hydroxynonenal,HNE)表达的影响。方法:采用单次双侧脑室注射Aβ蛋白的方法制备老年痴呆大鼠模型,分依达拉奉治疗组(1次/d,3 mg/kg,皮下注射)、空白对照组和模型组。自手术前1 d起给药,第10 d,各组分别进行Morris水迷宫检测后,处死动物并取脑,生物化学方法测定大鼠海马组织MDA含量,免疫组织化学技术检测海马HNE表达。结果:模型组与对照组相比水迷宫实验潜伏期时间显著延长,目的象限停留时间百分比降低,前脑组织MDA含量增加,海马CA1区HNE表达增多。依达拉奉组较模型组水迷宫结果显著改善,前脑组织MDA含量减少,且海马CA1区HNE表达显著减少。结论:依达拉奉能提高老年痴呆大鼠的学习和记忆能力,这种作用可能通过降低MDA及HNE等自由基氧化损伤产物而得以实现。

Abstract:Objective: To study the effects of Edaravone on learning and memory dificits,forebrain malondialdehyde(MDA) level and the expression of hippocampal 4-hydroxynonena(HNE) in rats with senile dementia.Methods: Senile dementia rat model was established by β-amyloid peptide bilateral ventricles injection.Three groups were set in the experiment: edaravone treatment group(subcutaneous injections,one time per day,3 mg/kg),model group and control group.Drug was administered from day 1 before operation.At day 10,after Morris water maze test was performed,the rats were killed and the brains were taken out.Biochemistry method was used to detect the level of MDA,and immunohistochemistry was used to observe the expression of HNE in hippocampus.Results: The latency was significantly increased in model group compared to control group in water maze experiment.The percentage of staying in objective quarant decreased.The level of MDA and the expression of HNE increased in forebrain and hippocampus CA1,respectively.In edaravone group,learning ability was greatly improved compared to model group.The level of MDA was decreased in forebrain and the expression of HNE was significantly decreased in hippocampus CA1.Conclusion: Edaravone improves the learning ability and memory in senile dementia rats,its effects may result from the decrease in hippocampal MDA level and HNE expression.
