摘    要:目的 探讨小鼠皮肤移植后IL-17蛋白及mRNA在外周血及移植物中的表达.方法建立三组小鼠皮肤移植动物模型:同基因组(BALBc→BALB/c)、异基因组(C57BL/6→BALB/c),脾细胞组(C57BL/6→BALB/c),每组40只.各组分别于移植后1、3、5、7d处死10只动物,采用ELISA法检测小鼠血清内IL-17水平,RT-PCR检测移植皮肤中的IL-17 mRNA的表达情况.结果 三组小鼠皮肤移植后第1天IL-17蛋白及mRNA表达有升高趋势,但差异无统计学意义(P>0.05).术后第3、5、7天异基因组IL-17蛋白及mRNA表达明显高于同基因组、脾细胞组(P<0.01),而后两组间差异无统计学意义(P>0.05).结论 IL-7在小鼠皮肤移植后早期发生急性排斥反应时呈高表达,有望作为预测和纠正排斥反应的观察指标.受体输注供体脾细胞可明显减轻异基因皮肤移植的排斥反应.

Abstract:Objective To investigate the expression of IL-17 and IL-17 mRNA in skin allograft and peripheral blood of recipient mice.Methods Three different models of skin grafting were perform in mice:the isotransplant group (BALB/c→BALB/c),the allotransplantation group (C57BL/6→BALB/c)and the latter plus donor spleen cells infusion group(C57BL/6→BAIB/c)(40 each).The concentration of IL-17 in peripheral blood plasma was measured by ELISA on the 1st,3rd,5th and 7th day after transplantation or infusion.and RT-PCR was performed to detect the expression of IL-17 mRNA in skin allograft at the same time points.Results There was no significant difference in the expressions of IL-17 and IL-17 mRNA in all the 3 groups of recipient mice one day after skin transplantation (P>0.05).On day 3,5 and 7 after skin graft transplantation,the expressions of IL-17 and IL-17 mRNA were lower in both isotransplant group and donor spleen cells infusion group than in allotrarlsplantation group,and the difference was statistically different(P<0.01);the expressions of IL-17 and IL-17 mRNA were lower in isotransplant group than in donor spleen cells infusion group, but the difference showed no statistical significance (P>0.05).Conclusions The high expression levels of IL-17 and IL-17 mRNA may be detected at the early acute rejection period in recipient mice.IL-17 may serve as a predictive and prognostic marker for acute rejection.Infusion of donor spleen cells to the recipients before skin graft transplantation can significantly alleviate rejection reaction of allografting.
