SDF-1在大肠腺癌中的表达及意义

【目的】探讨基质细胞衍生因子1(stromal cell-derived factor 1,SDF-1)在大肠腺癌发生、发展、浸润转移中的作用及临床意义。【方法】应用免疫组织化学(immunohistochemical,IHC)SP法检测90例大肠腺癌、45例非肿瘤性大肠黏膜组织中SDF-1的蛋白表达情况;应用逆转录聚合酶链法(RT-PCR)检测50例大肠腺癌、30例非肿瘤性大肠黏膜组织中SDF-1的mRNA表达情况。【结果】IHC检测结果:SDF-1蛋白在大肠腺癌中的阳性表达明显高于非肿瘤性大肠黏膜(P<0.01)。RT-PCR检测结果:SDF-1mRNA在大肠腺癌组中的表达量显著高于非肿瘤性大肠黏膜组(P<0.01)。大肠腺癌组中SDF-1蛋白、mRNA的表达量与癌组织的浸润深度、有无淋巴结转移有关(P<0.05),与性别、年龄、癌组织的分化程度无关(P>0.05)。【结论】SDF-1在大肠腺癌中高表达,且与癌组织的浸润深度、淋巴结转移有关,提示SDF-1可能与肿瘤的发生、浸润转移密切相关。

Expression and significance of SDF-1 in colorectal adenocarcinoma

【Objective】The purpose of the study was to discuss the role and clinical significance of SDF-1 in the development,infiltration and metastasis of colorectal carcinoma.【Methods】 The protein expression of SDF-1 was detected by SP immunohistochemical method in 90 cases of colorectal adenocarcinoma and 45 cases of normal colorectal mucosa;The expression of SDF-1 mRNA was detected by RT-PCR method in 50 cases of colorectal adenocarcinoma and 30 cases of normal colorectal mucosa.【Results】1.The SDF-1 protein positive expression and the expression quantity of SDF-1 mRNA in colorectal adenocarcinoma were respectively obviously higher than those in normal colorectal mucosa（P 0.01）.2.The SDF-1 protein and the expression quantity of SDF-1 mRNA were both correlated with the depth of tumor infiltration and metastasis of lymph node（P 0.05）,but they were not related with gender,age or differentiation of cancer（P 0.05）.【Conclusion】High expression of SDF-1 may play an important role in tumorigenesis,infiltration and metastasis of colorectal adenocarcinoma.
