巴戟天醇提物对离体大鼠缺血再灌注损伤心肌能量代谢的影响

摘    要:目的探讨巴戟天醇提物对缺血再灌注损伤(IRI)心肌能量代谢的影响。方法取健康雄性Wistar大鼠40只,随机均分为空白对照组、IRI组、巴戟天高剂量组和巴戟天低剂量组四组,每组10只,采用Langendorff灌流技术制备心肌IRI模型,2个巴戟天用药组分别在预灌时给予相应浓度的药物。灌流结束后,取各组大鼠左心室组织采用高效液相色谱法检测高能磷酸化合物。结果与IRI组相比,巴戟天高剂量组和巴戟天低剂量组三磷酸腺苷(ATP)、二磷酸腺苷(ADP)、一磷酸腺苷(AMP)、总腺嘌呤核苷酸含量(TAN)及心肌细胞能荷(EC)显著升高(P<0.05)。结论巴戟天醇提物能够改善IRI心肌的能量代谢,可能通过该途径发挥抗心肌IRI作用。

Abstract:Objective To investigate the influences of radix morindae officinalis ethanolie extracts on energy metabolism in rats with myocardial IRI. Methods 40 healthy male Wistar rats were randomly divided into 4 groups: the control group, the IRI group, the Radix morindae officinalis high dose group and the Radix morindae officinalis low dose group, and each group had 10 rats. A myocardial IRI model of rat was established with the Langendorff method, and all the medication groups were given with corresponding concentration drugs before perfusion. After perfusion, the high energy phosphates were de- termined by high performance liquid chromatography in myocardium of l0 rats in each group. Results Compared with the IRI group, the adenosine triphosphate (ATP), adenosine diphosphate (ADP), adenosine monophosphate (AMP), total adenine nucleotides (TAN) levels and the energy charge (EC) were increased significantly in the Radix morindae officinalis high dose group and the Radix morindae officinalis low dose group (P 〈 0.05). Conclusion Radix morindae officinalis ethanolic ex- tracts can improve energy metabolism of myocardium with IRI in rats, this may be the way to protect myocardium with IRI.
