内皮唾液酸蛋白在非小细胞肺癌中的表达及临床意义

摘    要:目的内皮唾液酸蛋白(CD248)是近年来发现的人类肿瘤标记物之一,在人类多种恶性肿瘤组织中异常高表达,并对肿瘤的细胞生长、血管生成、浸润及转移有重要作用。本研究通过检测CD248在非小细胞肺癌中的表达状态,希望揭示CD248与非小细胞肺癌的可能关系,探讨其与各临床病理因素的相关性。方法肿瘤组织经甲醛固定,石蜡包埋。免疫组化方法检测石蜡切片中CD248蛋白表达水平。结果CD248蛋白呈现棕褐色、细颗粒状物,弥漫性分布非小细胞肺癌细胞膜上和细胞间质,CD248蛋白在非小细胞肺癌组织中表达阳性率为73.08%。生物学统计结果表明,CD248蛋白表达与淋巴结转移及肿瘤临床分期等密切相关(P均<0.01);而与肿瘤患者的年龄和性别、肿瘤大小、病理类型及肿瘤部位等无关。结论CD248可能是一个参与非小细胞肺癌发生发展及浸润、转移有价值的分子标记物。

Abstract:Objective Endosialin(TEM 1/CD248)was recently identified as one of the tumor endothelial markers(TEMs).It's upregulated in most human cancers,and associated with the carcinogenesis,neovascularization,invasion and metastasis.So far,no research on CD248 in non-small-lung cancer has been reported.In this study we detected CD248 expression to explore the possible implication of CD248 in non-Small-lung cancer and its relation with caneer clinicopathological variables.Methods The expression of CD248 in 52 surgically resected specimens of non-small-lung cancer were determined by immunohistochemical method.The X 2 test was used.Results The CD248 proton was located in the membrane and intercellular substance of non-small-lung tumor cells with a positive rate of 73.08%.Biological statistical evaluation showed that the expression 0f CD248 protein had a close relation with lymph node metastasis and clinical stages(P<0.01),but was not related with age,gender,tumor size,histopathology type and tumor location in vivo.Conclusion CD248 is possibly a valuable molecular marker involved in NSLC carcinogenesis,invasion and metastasis.
