超声波辅助提取三七总黄酮工艺研究

摘    要:目的:优化超声波辅助提取三七中总黄酮含量的工艺.方法:以总黄酮提取率为考察目标,采用均匀设计试验法,重点考察甲醇提取液浓度、料液比、三七浸泡时间、超声时间4个因素对总黄酮提取率的影响.通过逐步回归分析及非线性二次规划得到最佳提取方案并进行验证.以最佳提取方案考察不同规格文山三七中总黄酮含量.结果:经逐步回归分析由甲醇提取液浓度和浸泡时间可以得到相关系数高达0.999 7的二次非线性回归方程.根据回归分析建立的方程,经非线性二次规划得到最佳提取条件:甲醇浓度20%,提取液加入比例60倍,三七浸泡时间20 min,超声时间5 min.通过验证表明此结果准确可信.经逐步回归分析发现甲醇浓度与浸泡时间对总黄酮提取率的影响较大,超声时间、料液比对总黄酮提取率影响很小;实验因素与总黄酮提取率之间呈现出典型的二次非线性回归.采用最佳提取工艺对春10头、春20头、春30头、春40头、春60头、春80头、春120头、冬40头,冬60头、冬120头(劣)共10个规格,126个文山三七样本中的总黄酮含量进行了测定,结果表明不同采收季节对三七品质影响不大.结论:三七品质越好,总黄酮含量越高,药效越好.

Abstract:Objective:The extraction of total flavonoids from panax notoginseng with ultrasonic wave was studied.Method:According to uniform experimental design,the influence of four parameters on the extraction rate of total flavonoids were investigated importantly, including methanol concentration,material/extraction solution ratio,immersing time and ultrasonic extraction time.Result: Through stepwise regression analysis,it was found that methanol concentration and immersing time had a deep influence to extraction rate of total flavonoids and ultrasonic extraction time and material/extraction solution ratio had relatively little influence.Furthermore, a classical 2nd order non-linear regression was found between four test parameters and extraction rate.The optimum extraction conditions Were as follows:methanol concentration 20%,material/extraction solution ratio 60,immersing time 20 min,ultrasonic extraction time 5 rain.Using this experimental design,10 standards of panax notoginseng,126 samples were studied.Conclusion:The results show that better panax notoginseng would include more flavonoids with better efficacy.
