C-myc反义寡核苷酸抗恶性黑素瘤作用机制初探

摘    要:目的 初步探讨C-myc反义寡核苷酸(ASODN)抗恶性黑素瘤可能作用机制。方法 以全硫代修饰的C myc反义寡核苷酸体外转染恶性黑素瘤细胞株A375,用台盼蓝活细胞计数法检测其对细胞增殖的影响,流式细胞仪检测细胞周期变化,间接免疫荧光流式细胞术检测HLA Ⅰ抗原的水平。结果 与对照组相比,①30μmol/LASODN组细胞生长明显受抑制,且发生G1 期阻滞;②ASODN组细胞上HLA Ⅰ抗原的水平明显上调。结论 C myc反义寡核苷酸可能通过抑制A375M细胞的增殖和上调HLA Ⅰ抗原水平的双重机制发挥抗肿瘤作用。

Abstract:Objective To investigate the possible mechanism of c-myc antisense oligodexynucleotide (ASODN) against malignant melanoma cells.Methods C-myc ASODN was synthesized and transfected into melanoma cell line A375. The viability of the cells was determined by the trypanblue exclusion test.Cell cycle was determined by flow cytometry.HLA-Ⅰexpression was examined by indirect immunofluorescence flow cytometry. Results The proliferation of A375 cells exposed to 30 μmol/L ASODN was decreased as compared to control group and SODN group.Cell cycles were blocked in G 1stage.The expression of HLA-Ⅰantigen was up-regulated by ASODN.Conclusion The inhibitory effect on malignant melanoma cells may be performed by the antiproliferation of c-myc ASODN and the up-regulation of HLA-Ⅰexpression by c-myc ASODN.
