摘    要:目的探讨食管癌瘤内注射生物可降解5-氟尿嘧啶聚乳酸纳米微粒(5-Fu-NPs)缓释剂所产生的抗肿瘤活性,为临床应用提供实验依据。方法36只SCID小鼠随机分为6组,分别是瘤内注射生理盐水的对照组、瘤内注射5-Fu-NPs(包括低剂量和高剂量)组、瘤内注射无载药NPs组、瘤内注射5-Fu组及腹腔内注射5-Fu组。观察荷瘤鼠肿瘤生长,荷瘤鼠给药前、后血象及给药后肿瘤组织的凋亡指数。结果瘤内注射5-Fu-NPs缓释剂组小鼠(20mg/kg体重,q3d×3)肿瘤生长缓慢,瘤体明显小于5-Fu腹腔注射给药组,并呈现良好的量效关系。结论瘤内注射5-Fu-NPs缓释剂的抗肿瘤效果优于局部和全身单纯5-Fu给药,它具有较重要研发价值。

Abstract:Objective To investigate the antitumor effects of biodegradable injectable gel of 5-flu- orouracil-loaded polyactic acid nanoparticles (5-Fu-NPs) on esophagus cancer nude mice and to further provide experimental foundation for clinical application.Methods Sixty SCID mice were randomized into six groups,individual tumor growth was observed,and drug related systemic toxicity was also evaluated after administration of 5-fluorouracil-loaded polyactic acid sustained-release preparation.The apoptotic in- dex (AI) by the terminal deoxynucleotidyl transterase-mediated dUTP biotin nick end labeling (TUNEL) technique on serial sections of formalin-fixed and paraffin-embedded tissues was measured.Re- suits The tumor volume of mice with intratumoral injection of 5-fluxrouracil-loaded polyactic acid sus- tained-release preparation was increased slower than that of those with intraperitoneal 5-fluorouracil,and exhibited dose-effective relationship.Conclusion The antitumor effect of intratumoral administration of 5-fluorouracil-loaded polyactic acid is better than that of local and systemic administration of 5-Fu alone.
